摘要
通过耐热试验以及牛津杯法从翡翠贻贝中分离筛选出具有良好抗菌活性的耐热海洋真菌PV-1。结合形态学与ITS测序确定菌株PV-1种属,采用单因素法进行培养基优化提升抗菌效果,用高效液相色谱法初步分析活性成分,通过测量菌落直径确定生理特性。PV-1经鉴定为土曲霉。实验表明,菌株PV-1对大肠杆菌(Escherichia coli)与吉氏库特氏菌SL-2(Kurthia gibsonii strain sp.)有良好的抗菌活性,经优化抑菌率分别提升至71.79%、71.52%,具有较强的抗菌活性,且富含极性与非极性活性成分;最适生长温度为35℃左右、盐度为1.5%~2.5%、pH值为6~7,具有兼性海洋特性,能在15~50℃、盐度0%~20%、pH值2~12条件下生长,具有极强的耐热、耐盐、耐酸碱能力,对环境耐受能力强,具有深入研究的价值。
Thermotolerant marine fungi with PV-1 good antibacterial activity were isolated and screened from Perna viridis by heat-resistant test and Oxford cup method. The species of the preferred strain PV-1 was determined by combining morphology and ITS sequencing. The single factor method was used to optimize the culture medium to improve the antibacterial effect, and the active components were preliminarily analyzed by high performance liquid chromatography, and the physiological characteristics were determined by the determination of colony diameter. After identification, PV-1 is Aspergillus terreus. Experiments show that strain PV-1 has good antibacterial activity against Escherichia coli and Kurthia gibsonii strain sp. to 71.79 % and 71.52 %, has strong antibacterial activity, and is rich in polar and non-polar active ingredients;the optimum growth temperature is about 35 ℃, salinity is 1.5 %~2.5 %, pH value is 6~7. It can grow under the conditions of 15~50 ℃,salinity 0 %~20 %, pH value 2~12, and has strong heat resistance, salt resistance, acid and alkali resistance and facultative marine characteristics. It has strong environmental tolerance and has the value of in-depth research.
