摘要
目的 探究针刺十四经脉治疗的特点和可能的分子机制。方法 基于针刺效应数据展示平台腧穴-靶点数据,利用Cytoscape3.8.2软件构建经脉-腧穴-靶点网络,并筛选核心靶点,采用Metascape数据库对核心靶点进行GO功能、KEGG通路富集分析和DisGeNET富集分析,使用DAVID6.8平台对十四经脉特异性靶点进行分析。结果 筛选得到十四经脉对应核心靶点19个,包括IL6、IL10、BDNF、NGF等。得到GO分析结果 573条,KEGG通路富集结果 99条,DisGeNET富集结果 20条。十四经脉特异性靶点分析显示,足阳明胃经特异性靶点有83个,包括ABCA1、CCL5、TIMP1、CCL2、RHEB等;督脉特异性靶点有50个,包括GFRA1、FRA1、TNFRSF11B、AGTR1、APLNR、CNR1等;手厥阴心包经特异性靶点有50个,包括ADORA2A、CYP2B6、ABCB1、GNAS、NLRP3等。结论 针刺十四经脉具有广谱效应,这可能与调节神经内分泌系统、神经免疫系统有关。同时,针刺十四经脉也具有特异效应,这可能与十四经脉的特异性靶点相关。
Objective To explore the molecular mechanism of acupuncture for the treatment of fourteen meridians. Methods Based on the acupoint-target data in acupuncture effect data display platform, Cytoscape 3.8.2was used to construct “meridian-acupoint-target” network, and the core targets were screened. Then Metascape database was used to analyze the GO function, KEGG pathway enrichment analysis and DisGeNET enrichment analysis of the core targets. Finally, the DAVID 6.8 platform was used to analyze the unique targets of the fourteen meridians. Results 19 core targets corresponding to the fourteen meridians were obtained, including IL6, IL10,BDNF, NGF, etc. 573 GO analysis results, 99 KEGG pathway enrichment results, and 20 DisGeNET enrichment results were obtained. The analysis of unique targets in the fourteen meridians showed that there were 83 specific targets in the ST, including ABCA1, CCL5, TIMP1, CCL2, RHEB, etc.;there were 50 unique targets in the GV,including GFRA1, FRA1, TNFRSF11B, AGTR1, APLNR, CNR1, etc.;there were 50 unique targets in the PC,including ADORA2A, CYP2B6, ABCB1, GNAS, NLRP3, etc. Conclusion Acupuncture on the fourteen meridians has a broad-spectrum effect, which may be related to the regulation of the neuroendocrine system and the regulation of the neuroimmune system. At the same time, acupuncture also has a unique effect, which may be related to the unique targets of the fourteen meridians.
