秦皮甲素通过下调FBI-1表达抑制人结直肠癌SW480细胞迁移和侵袭

Aesculin inhibits migration and invasion of human colorectal cancer SW480 cells by down regulating FBI-1 expression

摘要 目的:探讨秦皮甲素对人结直肠癌SW480细胞迁移和侵袭能力的影响,并研究其具体的分子机制.方法:以人结直肠癌SW480细胞作为研究对象,采用细胞划痕和Transwell实验检测细胞迁移和侵袭能力,采用shRNA干扰技术使SW480细胞中人类免疫缺陷病毒短转录诱导物连接因子-1(factor that binds to indeucer of short transcripts-1,FBI-1)基因的沉默,利用Western blot法检测波形蛋白(Vimentin)、E-钙黏蛋白(E-cadherin)和FBI-1等蛋白的表达变化.结果:该药物秦皮甲素可使人结直肠癌SW480细胞愈合能力、迁移和侵袭能力均显著下降,同时下调Vimentin和FBI-1蛋白表达水平以及上调E-cadherin蛋白表达水平,上述作用具有浓度依赖性.但经shRNA沉默FBI-1基因表达后,该蛋白的表达水平显著下降;同时FBI-1干扰SW480细胞中E-cadherin蛋白表达水平显著升高,Vimentin蛋白表达水平均显著下降,同时细胞迁移和侵袭能力也降低.结论:秦皮甲素具有浓度依赖性地抑制人结直肠癌SW480细胞迁移和侵袭能力,这可能与秦皮甲素抑制FBI-1蛋白表达,继而下调Vimentin蛋白表达和上调E-cadherin蛋白表达有关. Objective To investigate the effect of aesculin on the migration and invasion of human colorectal cancer SW480 cells and its specific molecular mechanism.Methods Human colorectal cancer SW480 cells were taken as the research object,and cell scratch and transwell assay were used to detect the ability of cell migration and invasion.SW480 cells were silenced by shRNA interference.The expressions of Vimentin,E-cadherin,and FBI-1 proteins were detected by western blot.Results The drug significantly reduced the healing ability,migration,and invasion ability of human colorectal cancer SW480 cells,and down-regulate the protein expression levels of Vimentin and FBI-1 and up-regulate the protein expression levels of E-cadherin.The above effects are significantly different between different concentrations.However,the expression level of the FBI-1 gene was significantly decreased after shRNA silencing.At the same time,the expression of E-cadherin protein and Vimentin protein in FBI-1 interfered SW480 cells were significantly increased,the expression of Vimentin protein was significantly decreased,and the migration and invasion ability of the cells was also decreased.Conclusion Aesculin can inhibit the migration and invasion of human colorectal cancer SW480 cells in a concentration-dependent manner,which may be related to the inhibition of FBI-1 protein expression by aesculin,and then down-regulate Vimentin and up-regulate E-cadherin protein expression.
