基于多元统计分析对茜草与藏茜草特征图谱的研究 被引量:2

Study on characteristic fingerprints of Rubiae Radix et Rhizoma and Rubia Wallichiana Radix et Rhizoma based on multivariate statistical analysis

摘要 目的:采用高效液相色谱法建立茜草、藏茜草(基源为光茎茜草和西藏茜草)的特征图谱,对茜草和藏茜草进行多元统计分析,为藏茜草的使用和基础研究提供参考。方法:采用ACE Excel 5C18(250 mm×4.6 mm,5μm)色谱柱,以乙腈-0.1%三氟乙酸(氨水调pH 2.6)为流动相,梯度洗脱,流速1.0 mL·min^(-1),检测波长270 nm,柱温30℃,采用Chempattern化学计量软件进行统计分析。结果:相似度分析表明茜草、光茎茜草和西藏茜草的特征图谱有明显不同;聚类分析将茜草和藏茜草聚为两大类,藏茜草中的光茎茜草和西藏茜草也聚为2类;主成分分析中各主要成分别为茜草中16号色谱峰的化合物和大叶茜草素,光茎茜草中14号色谱峰的化合物,西藏茜草中1号色谱峰的化合物。结论:利用建立的特征图谱和评价标准可鉴别茜草和藏茜草。 Objective:To establish an HPLC characteristic fingerprints of Rubiae Radix et Rhizoma and Rubia Wallichiana Radix et Rhizoma(the basic sources are Rubia wallichiana Decne.and Rubia tibetica Hook.f.).The multivariate statistical analysis of Rubiae Radix et Rhizoma and Rubia Wallichiana Radix et Rhizoma was carried out to provide reference for the use and basic research of Rubia Wallichiana Radix et Rhizoma.Methods:The HPLC analysis was carried out on an ACE Excel 5 C18column(250 mm×4.6 mm,5μm)with acetonitrile-0.1%trifluoroacetic acid(adjusting pH 2.6 with ammonia water)was used as the mobile phase.The flow rate was 1.0 mL·min^(-1),the detecion wavelength was 270 nm and the columntemperature was 30℃.Chempattern chemometrics software was used for statistical analysis.Results:Similarity analysis showed that the characteristics of Rubiae Radix et Rhizoma,Rubia wallichiana Decne.and Rubia tibetica Hook.f.were obviously different.Cluster analysis clustered Rubiae Radix et Rhizoma and Rubia wallichiana Radix et Rhizoma into two groups,and Rubia wallichiana Decne.and Rubia tibetica Hook.f.were also clustered into two groups.The main components in the principal component analysis was the compound of chromatographic peak No.16 and Mollugin in Rubiae Radix et Rhizoma,the compound of chromatographic peak No.14 in Rubia wallichiana Decne.,and the compound of chromatographic peak No.1 in Rubia tibetica Hook.f.,respectively.Conclusion:The established characteristic maps and evaluation criteria can be used to identify Rubiae Radix et Rhizoma and Rubia wallichiana Radix et Rhizoma.
