长链非编码RNA-PTENP1在结直肠癌组织中的表达及意义

Expression of long non-coding RNA-PTENP1 in colorectal cancer tissues

目的探讨长链非编码RNA-PTENP1在结直肠癌组织中的表达,以及与患者预后的相关性。方法选取2013年8月至2015年8月行治疗的结直肠癌患者106例,利用RT-qPCR技术检测组织中PTENP1的表达,于术后第1天开始对病例随访,截止时间2021年8月31日,所有病例随访满5年,以死亡作为结局事件,记录患者总生存时间和5年生存率。结果PTENP1在结直肠癌组织中的表达量低于癌旁组织,差异有统计学意义(P<0.001);PTENP1在不同分化程度、TNM分期、浸润深度、是否发生脉管浸润和淋巴结转移的结直肠癌患者组织中的表达量差异有统计学意义(P<0.05);低表达组患者平均生存时间和5年生存率低于高表达组(P<0.001);分化程度、TNM分期、淋巴结转移、PTENP1低表达是结直肠癌患者生存期的风险因素(HR=0.518、2.061、2.342和0.424,P<0.05)。结论PTENP1在结直肠癌患者组织中表达量降低,且与代表恶性进展的临床指标有关,与患者不良预后有关。

Objective To explore the expression of long non-coding RNA-PTENP1 in colorectal cancer(CRC)tissues,and its influence on the prognosis of patients.Methods CRC patients(n=106)who visited in our hospital(August 2013 to August 2015)were enrolled.The expression of PTENP1 in CRC tissues was detected by real-time polymerase chain reaction(RT-qPCR).The followed-up were performed for 5 years,from the first day following surgery until August 31,2021.The end-point events were death,and the overall survival time and 5-year survival rate of the patients were assessed.Results The expression of PTENP1 in CRC tissue was significantly lower than that in adjacent tissue(P<0.001).The expressions of PTENP1 were significant differences among patients with different histological grading,TNM stages,invasion depth,vascular invasion and lymph node metastasis(P<0.05).The average survival time and 5-year survival rate of patients with low expression were significantly lower than those of patients with high expression(P<0.001);histological grading,TNM stage,lymph node metastasis,and low expression of PTENP1 were risk factors for the survival of CRC patients(HR=0.518,2.061,2.342 and 0.424,P<0.05).Conclusion For CRC patients,the expression level of PTENP1 in the tissues reduced,which is related to clinical indicators representing malignant progression and to poor prognosis of patients.
