黄芩苷对环孢素致免疫功能低下小鼠免疫功能的影响 被引量:4

Effects of baicalin on immunologic function of immunocompromised mice induced by cyclosporin

摘要 目的探讨黄芩苷对环孢素所致免疫功能低下小鼠的免疫调节作用。方法将60只昆明小鼠随机分为正常对照组,模型组,阳性对照组,黄芩苷高、中、低剂量组,每组10只。除正常对照组外,其余5组均腹腔注射环孢素25 mg·kg^(-1)·d^(-1),隔日1次,共注射3次。造模成功后第2日起,正常对照组、模型组生理盐水连续灌胃,黄芩苷高、中、低剂量组黄芩苷溶液100、50、25 mg·kg^(-1)灌胃,阳性对照组贞芪扶正溶液1.76 g·kg^(-1)灌胃,均连续14 d。检测各组小鼠脾指数、胸腺指数,流式细胞仪检测外周血CD4^(+)、CD8^(+)T淋巴细胞百分比,ELISA法检测血清中白细胞介素(IL)-2、IL-4、IL-6、干扰素(IFN)-γ含量。结果与正常对照组比较,模型组脾指数和胸腺指数均显著下降(P<0.01),外周血中CD4^(+)T淋巴细胞百分比及CD4^(+)/CD8^(+)比值显著下降(P<0.01),血清中IL-2、IFN-γ浓度显著降低(P<0.01),IL-6浓度显著升高(P<0.01)。与模型组比较,黄芩苷高剂量组脾指数和胸腺指数显著升高(P<0.01),外周血中CD4^(+)T淋巴细胞百分比及CD4^(+)/CD8^(+)比值显著升高(P<0.05或P<0.01),IL-2、IFN-γ浓度显著上升,IL-6水平显著下降(P<0.01)。结论黄芩苷可以增强免疫功能低下小鼠的免疫功能。 AIM To investigate the immunomodulatory effect of baicalin on immunocompromised mice induced by cyclosporin.METHODS A total of 60 Kunming mice were randomly divided into normal control group,model group,positive control group,baicalin high,medium and low dose groups,with 10 mice in each group.Except the normal control group,the other 5 groups were given intraperitoneal injection of cyclosporin 25 mg·kg^(-1)·d^(-1)on alternate days for a total of 3 injections.From the second day after the successful modeling,the normal control group and the model group were continuously gavaged with saline,the baicalin high,medium and low dose groups were given baicalin solution by gavaged of 100,50 and 25 mg·kg^(-1)respectively,and the positive control group was gavaged with 1.76 g·kg^(-1)of Zhenqi Fuzheng solution for consecutive 14 d.The spleen and thymus indices of each group were measured.The percentage of CD4^(+)and CD8^(+)T lymphocytes in peripheral blood were measured by flow cytometry and the serum levels of IL-2,IL-4,IL-6 and IFN-γwere measured by ELISA.RESULTS Compared with the normal control group,the spleen index and thymus index in the model group decreased significantly(P<0.01),the percentage of CD4^(+)T lymphocytes and CD4^(+)/CD8^(+)ratio in peripheral blood decreased significantly(P<0.01),IL-2 and IFN-γconcentrations in serum decreased significantly(P<0.01),and IL-6 concentration increased significantly(P<0.01).Compared with the model group,the spleen index and thymus index in the baicalin high dose group increased significantly(P<0.01),the percentage of CD4^(+)T lymphocyte and CD4^(+)/CD8^(+)ratio in peripheral blood increased significantly(P<0.05 or P<0.01),the concentrations of IL-2 and IFN-γincreased and IL-6 levels decreased significantly(P<0.01).CONCLUSION Baicalin can enhance the immune function of immunocompromised mice.
