玉米ZmNAC28基因的克隆及其非生物逆境胁迫响应表达分析

摘要 NAC转录因子是植物特有的转录因子,在植物生长发育和胁迫响应中发挥重要作用。玉米基因组中有100多个NAC基因,但已完成功能解析的尚很少。本研究以玉米自交系W22为材料,利用RT-PCR技术克隆了一个NAC基因ZmNAC28,并对其生物信息学和非生物逆境胁迫下的表达水平进行分析。结果显示:ZmNAC28的编码区长909 bp,编码蛋白的N端有一个保守的NAC结构域,属于NAC转录因子家族成员;ZmNAC28在玉米根、茎、叶中表达水平高,其表达可被干旱、高盐和外源ABA诱导,并且该基因的启动子中存在多个非生物逆境胁迫响应元件。可见,ZmNAC28基因可能参与玉米对非生物逆境胁迫的响应,这为玉米NAC转录因子的功能解析奠定了基础。 NAC proteins are plant-specific transcription factors and play important roles in plant growth and development, and in stress responses. In the maize genome, more than 100 members of NAC genes have been identified, but almost all of them are not known for their functions in plants. In this study, we isolated a new NAC gene ZmNAC28 from maize inbred line W22 using reverse transcription PCR(RT-PCR), and analyzed its bioinformatics and expression under abiotic stresses. The results showed that the coding region of ZmNAC28 contains 909 base pairs. The ZmNAC28 protein encoded by ZmNAC28 gene had a conserved NAC domain in the N-terminal region, and was considered to be a member of NAC family. The ZmNAC28 gene was highly expressed in roots, stems and leaves in maize, and its expression could be induced by drought, high salt and exogenous abscisic acid(ABA). At the same time, multiple abiotic stress response elements were found in the promoter region of ZmNAC28. These results indicate that ZmNAC28 may be involved in the response process of abiotic stresses in maize, which lays a foundation for further revealing the function of NAC family members in maize.
