ACSL1在乳腺癌组织中的表达及对乳腺癌细胞侵袭影响与机制研究

摘要 目的探究长链脂酰辅酶A合成酶1(ACSL1)在乳腺癌组织中的表达及对乳腺癌细胞MDA-MB-231的侵袭影响与机制。方法收集蚌埠医学院第一附属医院肿瘤外科2021年10月至12月行乳腺癌根治手术获取的新鲜组织15例,qRT-PCR测定癌组织及配对的癌旁正常组织中ACSL1 mRNA表达差别,通过siRNA转染敲低乳腺癌细胞MDA-MB-231中ACSL1蛋白表达量,利用Western blot验明敲低效率。通过Transwell实验检测敲低ACSL1表达后MDA-MB-231细胞侵袭变化;采用Western blot测定PI3K/AKT信号通路蛋白表达量的变化。结果乳腺癌组织中ACSL1相对表达量较癌旁组织高(P<0.05)。敲低MDA-MB-231细胞ACSL1表达量后,细胞侵袭能力下降(P<0.05),且敲低ACSL1蛋白的乳腺癌细胞中p-PI3K与p-AKT蛋白相对表达量下调(P<0.05)。结论ACSL1在乳腺癌中表达显著升高,并且可能通过PI3K/AKT信号通路改变乳腺癌细胞MDA-MB-231的侵袭。 Objective To investigate the expression of ACSL1 in breast cancer tissues and its effect on invasion of breast cancer cells and its mechanism.Methods A total of 15 cases of fresh tissues from radical mastectomy in the Department of Surgical Oncology of the First Affiliated Hospital of Bengbu Medical College from October to December 2021 were collected.The difference of ACSL1 mRNA expression in cancer tissues and paired adjacent normal tissues was determined by qRT-PCR.The expression of ACSL1 protein in MDA-MB-231 breast cancer cells was knocked down by siRNA transfection,and the knockdown efficiency was verified by Western blot.The invasion of MDA-MB-231 cells after ACSL1 knockdown was detected by Transwell assay.The protein expression levels of PI3K/AKT signaling pathway were determined by Western blot.Results The relative expression level of ACSL1 in breast cancer tissues was higher than that in adjacent tissues(P<0.05).After knockdown of ACSL1 relative expression level in MDA-MB-231 cells,the cell invasion ability was decreased(P<0.05).In addition,the relative expression levels of p-PI3K and p-AKT were down-regulated in breast cancer cells with the knockdown of ACSL1 protein(P<0.05).Conclusion ACSL1 expression is significantly increased in breast cancer,and may affect the invasion of breast cancer cell mDA-MB-231 through PI3K/AKT signaling pathway.
