盐酸多西环素片在猪体内的残留消除规律研究

摘要 研究盐酸多西环素片按说明书给药后在猪体内的残留消除规律并确定休药期。将多西环素片以5 mg/kg体重口服给药,间隔24 h,连续给药5次。最后一次给药之后分别在第0日(12 h)、1日、3日、6日和8日时间点采集猪的肌肉、肝脏、肾脏和脂肪,采用建立并验证的HPLC-VWD法测定猪不同组织中多西环素的含量。方法学考察结果表明,在0.05-5μg/mL添加范围内的线性方程和相关系数为Y=0.044x-0.414,R2=0.999。不同组织中的盐酸多西环素的平均回收率在60.32%~116.80%。为保证兽药使用安全、食品安全与人民健康,建议按照休药期计算结果确定盐酸多西环素片在猪体内的休药期为7日。 In this study, it was determined the residual elimination rule of doxycycline hydrochloride tablets in pigs after administration according to the instructions and the drug withdrawal period. Doxycycline hydrochloride tablets were administered orally at 5 mg/kg of body weight, at an interval of 24 hours, for 5 consecutive doses. After the last administration, fat, muscle, liver and kidney were collected at 12 h, 1 d, 3 d, 6 d and 8 d respectively, and the content of doxycycline hydrochloride tablets in tissues was determined by HPLC-VWD. The linear range of doxycycline hydrochloride tablets in pig fat, muscle, liver and kidney tissues is 0.05-5 μg/ml. The linear equation and correlation coefficient are Y=0.044x-0.414, R~2=0.999. The average recovery rate of doxycycline hydrochloride in different tissues is 60.32%~116.80%. To ensure food safety and people’s health, it was suggested that the withdrawal time of doxycycline hydrochloride tablets in pig tissues should be no less than 7 days according to the calculation results.
