摘要
西藏色瓦绵羊是藏北特有草地型绵羊,由于自然交配情况下种公羊利用率低,需要利用人工授精方式来提高种公羊利用率以加快遗传改良进程,本研究旨在西藏色瓦绵羊上运用电刺激采精法寻求最佳采精刺激电压并结合按摩法以获得较好的色瓦绵羊种公羊人工采精效果。试验采用电刺激方法采集西藏色瓦绵羊种公羊精液,对采精时不同电压、按摩结合电刺激采精以及两次连续采精条件下所获得的精液进行品质检测。结果表明:电压在1~3 V和9~15 V时西藏色瓦绵羊种公羊无法采集到精液,在电压为5 V和7 V时公羊阴茎勃起并射精但其射精量、精子密度、精子活力无显著性差异(P>0.05);并且发现单纯使用电刺激和结合按摩所采集出的精液在精液量、精子密度、活力无显著性差异(P>0.05),但是射精时间有显著差异(P<0.05);在间隔30 min进行两次连续采精时第1次采精的采精量、精子密度均显著优于第2次的(P<0.05),活力无显著性差异(P>0.05)。说明西藏色瓦绵羊公羊电刺激采精时适宜电压为5~7 V,电刺激结合按摩可以有效缩短射精时间,但不宜短时间内连续采精,因此电刺激法可以在西藏色瓦绵羊人工授精时推广应用。
The Tibetan Seva sheep is an endemic grass-type sheep in northern Tibet.Due to the low utilisation rate of rams in natural mating,artificial insemination is needed to improve the utilisation rate of rams to speed up the genetic improvement process.The aim of this study is to find the optimal sperm collection stimulation voltage for Tibetan Seva sheep and to combine it with massage to obtain better artificial semen collection results for Seva sheep rams.The test was carried out to collect semen from Tibetan Seva rams using electrical stimulation and to test the quality of semen obtained under different voltages,massage combined with electrical stimulation and under two successive collection conditions.The results showed that the rams were unable to collect semen at voltages of 1~3 V and 9~15 V.The rams were erect and ejaculated at voltages of 5 V and 7 V,but there was no significant difference in ejaculate volume,sperm density and sperm vitality(P>0.05).In the case of the first collection,semen volume and sperm density were significantly better than those of the second collection(P<0.05)when two consecutive collections were performed at 30 min intervals,but there was no significant difference in viability(P>0.05).This suggests that the appropriate voltage for electrically stimulated sperm collection in Tibetan sheep rams is 5-7 V,and electric stimulation combined with massage can effectively shorten the ejaculation time,but is not suitable for continuous sperm collection in a short period of time.
