摘要
采用TTC法测定葛根种子生活力,通过单因素试验和Box-Benhnken响应面试验,探究染色时间、TTC浓度和染色温度对TTC染色的影响。结果表明,葛根种子TTC法染色的最佳条件为:染色时间2 h、TTC浓度0.57%、染色温度37.3℃,在优化条件下测得葛根种子平均生活力为84.44%,实际测定值与理论预测值误差为3.71%(<5%),说明响应面法优化葛根种子TTC法测定条件是可行的。
The seed viability was determined by TTC method,and the effects of dyeing time,TTC concentration and dyeing temperature on TTC dyeing were investigated by single factor test and Box-Benhnken response surface test.The results showed that the optimal conditions for TTC dyeing of pueraria seeds were as follows:dyeing time for 2 hours,TTC concentration of 0.57%,dyeing temperature at 37.3℃,the average viability of pueraria seeds was measured to be 84.44%under the optimized conditions.The error of the predicted value was 3.71%(<5%),indicating that the response surface methodology was feasible to optimize the TTC determination conditions of pueraria seeds.
