醋鸡内金配方颗粒的特征图谱建立及指标成分含量测定

Establishment of Characteristic Chromatogram Combined with Content Determination of Target Components in Vinegar-Processed Gallus Gallus Domesticus Formula Granules

目的:建立醋鸡内金配方颗粒特征图谱及指标成分含量测定方法。方法:采用ZORBAX Eclipse Plus C 18(250 mm×4.6 mm,5μm)色谱柱;以甲醇-0.2%磷酸水溶液为流动相,梯度洗脱;体积流量为1.0 mL/min→0.5 mL/min→0.8 mL/min→1.0 mL/min→1.0 mL/min;柱温为35℃;检测波长为254 nm。运用《中药色谱指纹图谱相似度评价系统(2012版)》软件对样品进行相似度评价,并确定共有峰。结果:建立的醋鸡内金配方颗粒的特征图谱及指标成分含量测定方法符合方法学验证要求。尿苷和鸟苷的进样量分别在0.0140~0.2799μg、0.0106~0.2121μg范围内有良好的线性关系。平均回收率分别为98.3%、103.1%。测定的7批醋鸡内金配方颗粒特征图谱相似度均>0.900,共标定了9个特征峰,并指认出其中4个成分,分别为:尿嘧啶、尿苷、腺苷和鸟苷。结论:该方法简便,准确,稳定性好,可用于醋鸡内金配方颗粒的质量控制。

Objective To establish a method for characteristic chromatogram and target components determination of Vinegar-processed Gallus gallus domesticus formula granules.Methods The determination was performed on Agilent ZORBAX Eclipse Plus C18(250 mm×4.6 mm,5μm)column;the gradient elution solvent system was composed of methanol-0.2%phosphoric acid at the flow rate of 1.0 mL/min→0.5 mL/min→0.8 mL/min→1.0 mL/min→1.0 mL/min;the column temperature was set at 35℃,and detection wavelength was set at 254 nm.The Similarity Evaluation System for Chromatographic Fingerprint of TCM(2012 edition)was used to evaluate the similarity and to confirm common peaks.Results The validation results showed that the method could be used for characteristic chromatogram and target components determination of vinegar-processed Gallus gallus domesticus formula granules.Uridine and guanosine had a good linearity within the range of 0.0140-0.2799μg,0.0106-0.2121μg,respectively.The average recovery of uridine and guanosine was 98.3%,103.1%,respectively.Nine common peaks were identified with similarity more than 0.900 in 7 batches of vinegar-processed Gallus gallus domesticus formula granules,and four of those were identified as uracil,uridine,adenosine,guanosine.Conclusion This simple method has good repeatability and stability,which can be used for quality evaluation of Vinegar-processed Gallus gallus domesticus formula granules.
