摘要
目的:探究丁酸钠(NaB)联合替莫唑胺(TMZ)对胶质瘤细胞凋亡及焦亡的影响。方法:将U251细胞分为空白对照组(无处理)、100μmol/L TMZ组、10 mmol/L NaB组及两者联合处理组。处理24 h后,采用细胞划痕实验及Transwell实验测定细胞的迁移能力及侵袭能力,采用Annexin V-FITC/PI双染法检测细胞凋亡率,DCFH-DA标记法检测细胞中活性氧水平,Western blot法检测细胞中焦亡相关蛋白消皮素(GSDMD)、GSDMD-N、NLRP3和IL-1β蛋白的表达。结果:NaB及TMZ均可抑制U251细胞的迁移和侵袭能力,促进细胞凋亡和活性氧生成(P<0.05),两者联合作用更强。TMZ与NaB均能促进U251中GSDMD蛋白的解离及IL-1β和NLRP3蛋白的表达,两者联合作用更强(P<0.05)。结论:NaB联合TMZ可以促进胶质瘤细胞的凋亡与焦亡。
Aim:To explore the effects of sodium butyrate and temozolomide on apoptosis and pyroptosis of glioma cells.Methods:U251 cells were divided into blank control group(no treatment),100 μmol/L temozolomide group, 10 mmol/L sodium butyrate group and combined treatment group.After 24 hours treatment, the migration capacity and invasion capacity of U251 cells were measured by cell scratch test and Transwell test, respectively.Apoptosis and reactive oxygen species(ROS) levels of U251 cells were detected by Annexin V-FITC/PI staining and DCFH-DA marking, respectively.The apoptosis related proteins GSDMD,GSDMD-N,NLRP3 and IL-1β in U251 cells were detected by Western blot.Results:Both sodium butyrate and temozolomide could inhibit the migration and invasion, promote apoptosis and ROS production, the dissociation of GSDMD protein, and IL-1β and NLRP3 expression of U251 cells, and the combined action was greater(P<0.05).Conclusion:The combined action of sodium butyrate and temozolomide can promote the apoptosis and pyroptosis of glioma cells.
