Application of organoid in the precision treatment of breast cancer

摘要 目的总结类器官在乳腺癌精准治疗中的应用,并探讨其发展方向。方法以“类器官、乳腺癌、精准治疗、个体化治疗”为中文关键词,以“organoids,breast cancer、precision treatment、personalized treatment”为英文关键词,应用PubMed及中国知网数据库检索2009年1月-2022年2月发表的相关文献。共检索到中文文献23条,英文文献337条。纳入标准:(1)类器官培养的进展;(2)类器官在乳腺癌精准治疗中的研究;(3)类器官研究面临的问题。排除标准:重复性、数据陈旧的相关文献。最终纳入57篇文献进行分析。结果乳腺癌类器官在基因编辑、耐药性验证、药物筛选及新药研发等方面取得有价值的进展,为临床研究提供参考。结论类器官培养技术具有缺陷性,缺乏多种细胞类型及完整免疫微环境成分,需探索新的培养方式完善此模型。 Objective To review the application of organoids in the precision treatment of breast cancer,and to explore its development direction.Methods PubMed and China National Knowledge Infrastructure(CNKI)database retrieval systems were used with"organoids,breast cancer,precision treatment,personalized treatment"as keywords from January 2009to February 2022.A total of 23Chinese literatures and 337English literatures were retrieved.Inclusion criteria:(1)advances in organoids culture;(2)studies of organoids in the precision treatment of breast cancer;(3)problems facing organoids research.Exclusion criteria included repetitive,relevant literature with stale data.Finally,57articles were included for analysis.Results Valuable progress has been made in gene editing,drug resistance verification,drug screening and new drug development of breast cancer organoids,which will provide reference for clinical research.Conclusion The organoids culture technique is defective,lacking multiple cell types and complete immune microenvironment components,so it is necessary to explore new culture methods to improve this model.
