摘要
目的:在胶体金免疫层析技术的基础上采用竞争法制备喹乙醇快速检测试纸条。方法:采用一锅法合成显色用的胶体金,调控柠檬酸钠的量,获得不同最大吸收波长的胶体金。通过对胶体金吸收波长、碳酸钾添加量等条件的优化,获得胶体金的最大吸收波长为522 nm、碳酸钾(0.2 mmol/L)添加量为15μL的最佳条件,在此最优条件下制备试纸条,考察试纸条的方法学参数,并将制备好的试纸条检测实际样品。结果:随着喹乙醇浓度的减小(5.3×10^(-1)mg/m L,5.3×10^(-7)mg/m L),试纸条的控制线C线一直显色,而检测线T线逐渐清晰、显色,表明试纸条是准确的,检出限为5.3×10^(-7)mg/m L,所有的检测在5~8分钟内能完成,并成功用于鲫鱼实际样品的检测。结论:该试纸条适用于对肉类中的喹乙醇残留进行高效灵敏的检测,有利于保障人们的食品安全。
Objective:On the basis of colloidal gold immunochromatography technology,competitive method was used to prepare rapid test strips for olaquindox.Methods:The colloidal gold for color development was synthesized by one pot method,and the amount of sodium citrate was adjusted to obtain colloidal gold with different maximum absorption wavelengths.By optimizing the absorption wavelength of colloidal gold and the addition amount of potassium carbonate,the maximum absorption wavelength of colloidal gold was522 nm and the addition amount of potassium carbonate (0.2 mmol/L) was 15μL,prepare the test strip under this optimal condition,investigate the methodological parameters of the test strip,and test the actual sample with the prepared test strip.Results:With the decrease of olaquindox concentration (5.3×10^(-1)mg/m L,5.3×10^(-7)mg/m L),the control line C of the test strip showed color all the time,while the detection line T was gradually clear and colored,indicating that the test strip was accurate,and the detection limit was 5.3×10^(-7)mg/m L,all tests could be completed within 5 to 8 minutes,and successfully used for the detection of actual samples of crucian carp.Conclusion:The test strip is suitable for the highly efficient and sensitive detection of olaquindox residues in meat,which is conducive to ensuring the safety and health of human food.
