中药骨痹通消颗粒通过调控Wnt/β-catenin通路治疗激素性股骨头坏死的实验研究 被引量:7

摘要 目的通过动物实验探讨中药骨痹通消颗粒通过调控Wnt/β-catenin通路治疗激素性股骨头坏死的价值。方法将造模成功的激素性股骨头坏死大白兔随机分为3组模型组、碳酸钙组与中药组,每组13只。模型组用生理盐水灌胃治疗,碳酸钙组碳酸钙D3颗粒治疗,中药组给予骨痹通消颗粒治疗。记录实验动物的变化情况与股骨头Wnt/β-catenin蛋白表达水平变化情况。结果碳酸钙组与中药组给药后6周、8周的体质量高于模型组,中药组高于碳酸钙组,对比差异都有统计学意义(P<0.05)。碳酸钙组与中药组给药后6周、8周血清超氧化物歧化酶(superoxide dismutase,SOD)含量高于模型组,丙二醛(malondialdehyde,MDA)含量低于模型组,对比差异有统计学意义(P<0.05),碳酸钙组与中药组对比差异也有统计学意义(P<0.05)。碳酸钙组与中药组给药后8周的股骨应变能力、最大载荷都高于模型组,中药组高于碳酸钙组,对比差异都有统计学意义(P<0.05)。碳酸钙组与中药组给药后8周的股骨Wnt、β-catenin蛋白相对表达水平高于模型组,中药组高于碳酸钙组,对比差异都有统计学意义(P<0.05)。模型组镜下见软骨细胞减少、成骨细胞减少,骨细胞固缩或消失;碳酸钙组骨小梁出现骨质紊乱、扭曲、毛糙,髓腔内脂肪细胞体积增大;中药组股骨头软骨细胞数量正常,排列规则,空骨陷窝较少。结论中药骨痹通消颗粒在激素性股骨头坏死实验动物中的应用能激活Wnt/β-catenin通路,改善股骨的生物力学状况,有利于维持正常机体氧自由基产生与清除处于平衡状态,从而发挥积极的治疗作用。 Objective To explore the values of Gubi Tongxiao Granules(骨痹通消颗粒)treating steroid-induced necrosis of femoral head by regulating Wnt/β-catenin pathway through animal experiments.Methods The rabbits with steroid-induced necrosis of femoral head successfully modeled were randomly divided into three groups:model group,calcium carbonate group and traditional Chinese medicine group,13 cases in each group.The model group were treated with saline in the stomach,the calcium carbonate group were treated with calcium carbonate D3 particles,and the traditional Chinese medicine group were treated with Gubi Tongxiao Granules.The changes of experimental animals and the expression of Wnt/β-catenin protein in femoral head were recorded.Results The weight of the calcium carbonate group and the traditional Chinese medicine group were higher than that of the model group at 6 and 8 weeks after administration,and that of the traditional Chinese medicine group was higher than that of the calcium carbonate group,and the differences were statistically significant(P<0.05).The serum SOD content of the calcium carbonate group and the traditional Chinese medicine group were higher than that of the model group at 6 and 8 weeks after administration,and the MDA content was lower than that of the model group and the differences were statistically significant(P<0.05),there were also statistically significant differences among the groups(P<0.05).The femoral strain capacity and maximum load of the calcium carbonate group and the traditional Chinese medicine group were higher than that of the model group,and that of the traditional Chinese medicine group was higher than that of the calcium carbonate group at 8 weeks after administration and the differences were statistically significant(P<0.05).The relative expression levels of Wnt and β-catenin protein in the femoral bone 8 weeks after administration of the calcium carbonate group and the traditional Chinese medicine group were higher than those in the model group,and those of the traditional Chinese medicine group were higher than those of the calcium carbonate group and the differences were statistically significant(P<0.05).In the model group,chondrocytes decreased,osteoblasts decreased and osteocytes contracted or disappeared.Bone trabeculae in the calcium carbonate group showed osteodystrophy,distortion and roughness,and the volume of fat cells in the medullary cavity increased.The numbers of bone chondrocytes were normal in the traditional Chinese medicine group,the arrangements were regular,and there were fewer empty bone pits.Conclusion The application of Gubi Tongxiao Granules in steroid-induced necrosis of femoral head experimental animals can activate the Wnt/β-catenin pathway,improve the biomechanical status of the femur,and help to maintain the normal body's oxygen free radical production and clearance in balanced state.
