糖皮质激素相关信号通路在阿尔茨海默病发生、发展中的作用及其研究进展

The role of the glucocorticoid⁃related signaling pathway and its research advances in the development and progression of Al⁃zheimer's disease

摘要 阿尔茨海默病(Alzheimer's disease,AD)是一种常见的神经退行性疾病,但其发病机制迄今仍未完全明了。有研究发现糖皮质激素(corticosteroids,GC)可诱发或促进AD的发生、发展。文章着重介绍了GC介导多种信号通路抑制Tau蛋白降解或促进Tau蛋白过度磷酸化等来促进AD的发生、发展。AD的病理表现主要有β-淀粉样蛋白(amyloidβ-protein,Aβ)形成的斑块和Tau蛋白的过度和异常磷酸化引起的神经纤维缠结(nerve fiber tangles,NFT)。文章旨在为调控GC代谢以预防或早期干预AD提供理论基础。 Alzheimer's disease(AD)is the common neurodegenerative disease.But its pathogenesis is still not fully under⁃stood.Some studies have found that corticosteroids(GC)can induce or promote the occurrence and development of AD.This paper fo⁃cuses on the GC⁃mediated signaling pathways that inhibit Tau protein degradation or promote Tau protein hyperphosphorylation to pro⁃mote the occurrence and development of AD.The pathological manifestations of AD mainly include plaques formed by amyloidβ⁃pro⁃tein(Aβ)and nerve fiber tangles(NFT)caused by excessive and abnormal phosphorylation of the Tau.This paper aims to provide a the⁃oretical basis for regulating GC metabolism to prevent or intervene AD in an early stage.
