基于六角锥体模型颜色直方图的植物农杆菌基因瞬时转化表达图像定量分析

摘要 基因的瞬时表达是一种快速的研究基因表达、蛋白质亚细胞定位及基因间相互作用的手段,有简单、快速、周期短等优点,结果直观可靠,但在分析实验结果时,运用肉眼观察法,对结果衡量的标准不统一,有一定的局限性.基于颜色恒常性,利用图像分割的阈值法对农杆菌外源基因侵染4种植物叶片的注射实验得到的图像进行分析,将植物基因瞬时转化表达的图像转为六角锥体模型颜色空间,进行颜色直方图的分析,研究表明此方法定量分析植物对外源基因瞬时转化表达更具实用性和科学性. Transient gene expression,having the advantages of simplicity and being a fast and short cycle,is a rapid method to study gene expression,protein subcellular localization and gene interaction.The results are intuitive but reliable,while in the analysis of experimental results,the visual observation method is often used to measure the results to see if the standard is not unified,and there are certain limitations with it.Based on color constancy and using the method of threshold image segmentation of agrobacterium exogenous gene in detail for four kinds of plant leaves,with injection images obtained by experimental analysis,plant transient expression images were transferred into hue-saturation-value color space;the color histogram analysis and quantitative analysis results showed that the method can determine exogenous gene expression in a more practical and scientific manner.
