铁死亡在术后认知功能障碍中的作用

Roles of ferroptosis in postoperative cognitive dysfunction

摘要 术后认知功能障碍(postoperative cognitive dysfunction,POCD)是老年患者术后常见的中枢系统并发症,表现为精神错乱、人格改变和记忆减退等,确切机制不详。铁死亡是一种新发现的调节性细胞死亡方式(regulated cell death,RCD),由铁依赖的脂质过氧化引起,在肿瘤和器官损伤中发挥重要作用。围术期的炎症反应、氧化应激及代谢异常都能导致神经细胞铁死亡。本文概括了铁死亡的发生机制,重点从炎症的角度探讨了铁死亡与POCD的关联,为预防和治疗POCD提供新思路。 Postoperative cognitive dysfunction(POCD) is one of common central nervous system complications after operation for aged patients,manifested as insanity,personality changes and hypomnesia,but the exact mechanism is unknown.Ferroptosis is a newly identified form of regulated cell death(RCD)caused by iron-dependent lipid peroxidation and plays an important role in tumor and organ damage.It was found that inflammation,oxidative stress and metabolic disorder could induce ferroptosis of nerve cells during perioperative period.This paper summarizes the mechanism of ferroptosis and focuses on the relationship between ferroptosis and POCD from the perspective of inflammation,which may provide new ideas for the prevention and treatment of POCD.
