摘要
为满足当前公安工作需要,建立一种操作步骤简化,可有效的去除PCR抑制剂,同时适用于大批量样本自动化操作的DNA检测方法。通过严格的磁珠筛选和优化提取试剂,建立微量DNA提取模型,比较不同提取试剂盒提取DNA的回收率、稳定性、灵敏度和抗抑制的能力,测试新型纳米磁珠试剂盒对案件不同生物检材的检出情况。结果表明,新型高效纳米磁珠试剂盒提取DNA操作步骤简单,可有效去除抑制剂而获取高质量DNA模板。新型高效纳米磁珠试剂盒提取效果优于目前公安检验中常用的试剂盒,有利于微量生物检材DNA检验。该试剂盒适用于自动化平台进行批量生物样本处理,可满足目前公安实际案件检验需要。
To meet the needs of current public security work, it is necessary to establish a DNA test method that can simplify the operation steps, effectively remove the PCR inhibitors, and is suitable for automatic operation of large quantities of samples. Through strict magnetic bead screening and optimization of extraction reagents, a micro DNA extraction model was established to compare the recovery rate, stability,sensitivity and anti-inhibition ability of different extraction kits for DNA extraction, and test the detection of new nano magnetic bead kits for different biological test materials in the case. The results showed that the new efficient nano magnetic bead kit was simple in DNA extraction and could effectively remove inhibitors to obtain high-quality DNA templates. The extraction effect of the new efficient nano magnetic bead kit is better than that of the kit commonly used in public security inspection at present, which is conducive to DNA detection of micro biological test materials. This kit is suitable for batch biological sample processing on the automated platform, and can meet the current needs of actual case inspection of public security.
