摘要
目的:探讨间歇有氧运动对大鼠心肌NOX4、ROS表达及血清相关氧化因子的影响.方法:选取雄性SD大鼠20只,随机分为对照组(C组)和间歇有氧运动组(CE组),每组10只.C组大鼠正常喂养,CE组大鼠进行为期4周跑台运动.训练结束后HE染色观察心肌组织形态结构,血流动力学测定各组大鼠心功能,紫外分光光度法测定血清MDA含量和SOD活性,微量酶标法测定血清LDH活性,DHE法检测心肌活性氧(ROS)水平,Western Blot检测心肌NOX4表达.结果:与C组比较,CE组大鼠心肌形态结构显著改善,LVEDP显著降低(P<0.01),LVSP和±dp/dtmax显著升高(P<0.01),血清MDA含量、LDH活性均显著降低(P<0.01),SOD活性显著增强(P<0.01),ROS水平均显著降低(P<0.01)心肌NOX4的表达显著降低(P<0.01).结论:间歇有氧运动可显著改善大鼠心脏形态结构,提升大鼠心功能;间歇有氧运动可使大鼠血清MDA、LDH的含量显著降低,SOD活性显著增强,心肌NOX4和ROS表达显著降低,进而降低大鼠心脏氧化应激水平,改善大鼠心功能.
To investigate the effects of intermittent aerobic exercise on the expression of NOX4,ROS and serum-related oxidative factors in rat myocardium.Methods:Twenty male SD rats were selected and randomly divided into control group(group C)and intermittent aerobic exercise group(group CE),10 rats in each group.The rats in group C were fed normally,and the rats in group CE were subjected to treadmill exercise for 4 weeks.After training,HE staining was used to observe the morphological structure of myocardial tissue,hemodynamics was used to measure the cardiac function of the rats in each group,serum MDA content and SOD activity were measured by UV spectrophotometry,serum LDH activity was measured by micro-enzyme labeling method,and myocardial reactive oxygen species was measured by DHE method.(ROS)level,and Western blot detection of myocardial NOX4 expression.Results:Compared with the C group,the myocardial morphology and structure of the CE group were significantly improved,the LVEDP was significantly decreased(P<0.01),the LVSP and±dp/dtmax were significantly increased(P<0.01),the serum MDA content and LDH activity were significantly decreased(P<0.01),SOD activity was significantly enhanced(P<0.01),ROS levels were significantly decreased(P<0.01)and myocardial NOX4 expression was significantly decreased(P<0.01).Conclusion:Intermittent aerobic exercise can significantly improve the morphological structure of the rat heart and improve the cardiac function of the rat;intermittent aerobic exercise can significantly reduce the serum MDA and LDH contents,significantly enhance the SOD activity,and significantly reduce the myocardial NOX4 and ROS expressions.,thereby reducing the level of oxidative stress in the rat heart and improving the cardiac function of the rat.
