摘要
目的 探讨心磷脂酰基转移酶1(LCLAT1)在喉鳞癌中的表达及其与预后的关系。方法 对3例喉鳞癌患者的肿瘤、癌旁及淋巴转移组织进行单细胞RNA测序。使用R软件对上述样本进行数据分析,观察LCLAT1在上皮细胞亚群中的表达情况。然后使用癌症基因组图谱(TCGA)中喉鳞癌的数据,分析了包含LCLAT1在内128个甘油磷脂合成途径相关基因的表达情况及对预后的影响,并使用Cytoscape软件评估了LCLAT1在喉鳞癌甘油磷脂合成途径中的重要作用。最后使用免疫组化实验检测LCLAT1在20例喉鳞癌标本中的表达情况。结果 识别出10个主要的细胞簇,其中上皮细胞包含11个亚群,标记为c0~c10。簇c1是处在高增殖状态的恶性细胞,并且高表达LCLAT1。LCLAT1作为甘油磷脂合成途径中的关键基因之一在肿瘤组织中高表达,并且是喉鳞癌的危险因素和独立预后指标。免疫组化验证LCLAT1在喉鳞癌肿瘤组织中表达量高于癌旁组织。结论 LCLAT1在喉鳞癌高增殖状态的恶性上皮细胞中高表达,可能作为甘油磷脂合成途径中的关键基因促进喉癌发展,影响喉癌预后。
Objective To investigate the expression of lysocardiolipin acy ltransferase I(LCLAT1)in laryngeal carcinoma and its correlation with prognosis.Methods Single cell RNA sequencing was performed in tumor,para-neoplastic and lymphatic metastasis tssues of three patients with laryngeal squamous cell carcinoma.R software was used to analyze the data of the above samples and the expression of LCLATI in epithelial cell subsets was observed.Then the data of laryngeal squamous cell carcinoma was used in The Cancer Genome Atlas(TCGA)to analyze the expression of 128 genes related to glycerophospholipid synthesis pathway,including LCLAT1,and the impact on prognosis.And then the important role of LCLAT1 in the glycerophospholipid synthesis pathway of laryngeal squamous cell carcinoma was evaluated by Cytoscape software.Finally,the expression of LCLAT1 in 20 laryngeal squamous cell carcinoma specimens was detected by immunohistochemistry.Results Ten major cell clusters were identified,including 11 subsets of epithelial cells labeled c0-c 10.Cluster cl was a malignant cell with high proliferation and high expression of LCLATI.As one of the key genes in the glycerophospholipid synthesis pathway,LCLATI was highly expressed in tumor tissues and was a risk factor and independent prognostic indicator of laryngeal squamous cell carcinoma.Immunohistochemistry confirmed that the expression ofLCLAT1 in laryngeal squamous cell carcinoma was higher than that in adjacent tissues.Conclusions LCLATI is highly expressed in the highly proliferating malignant epithelial cllof laryngeal squamous cell carcinoma,which may be a key gene in the glycerophospholipid synthesis pat-hway to promote the development of laryngeal carcinoma and afect the prognosis of laryngeal carcinoma.
