摘要
目的对Zeste增强子同源物2(EZH2)在脑胶质瘤中的最新研究进展进行汇总,介绍在脑胶质瘤中EZH2与非编码RNA相互作用机制及功能,为开发以EZH2为靶点的脑胶质瘤新型治疗方法提供参考。方法应用PubMed及CNKI期刊全文数据库检索系统,以“EZH2,脑胶质瘤,非编码RNA,表观遗传修饰”为中文关键词,以“EZH2,glioma,non-coding RNA,epigenetic modification”为英文关键词,检索2000-2022年发表的相关中英文文献。纳入标准:(1)EZH2在脑胶质瘤发生发展中的作用;(2)EZH2与非编码RNA调控脑胶质瘤的最新进展。排除标准:陈旧及重复的文献、研究结果不明确及不真实可靠的文献。最终共纳入符合条件文献87篇。结果EZH2可以促进脑胶质瘤细胞的生长、侵袭和转移、代谢等。EZH2可作为非编码RNA如miRNA和lncRNA的上游基因来抑制miRNA和lncRNA表达,从而促进脑胶质瘤的发生发展。此外,EZH2还受到miRNA、lncRNA和circRNA的调控,即EZH2也可作为非编码RNA的下游靶基因来调控脑胶质瘤的恶性进展。结论EZH2与非编码RNA构成复杂的调控网络在脑胶质瘤的发生发展中起重要作用,提示EZH2是脑胶质瘤的潜在治疗靶点。因此进一步研究EZH2在脑胶质瘤发生发展中的分子机制和功能,有助于开发以EZH2为靶点的新型脑胶质瘤治疗方案,以提高脑胶质瘤患者的治愈率。
Objective To summarize the latest research progress of Zeste enhancer homolog 2(EZH2)in brain glioma,introduce the interaction mechanism and function of EZH2 and non-coding RNA in brain glioma,and provide a reference for the development of new therapeutic methods for brain glioma targeting EZH2.Methods The PubMed and CNKI journal full-text database retrieval systems were used to search the relevant Chinese and English literatures published from 2000 to 2022 with"EZH2,glioma,non coding RNA,and epigenetic modification"as the Chinese key words and"EZH2,glioma,non-coding RNA,and epigenetic modification"as the English key words.Inclusive criteria:(1)The role of EZH2 in the occurrence and development of brain glioma;(2)Recent progress in the regulation of brain glioma by EZH2 and non-coding RNA.Exclusion criteria contained obsolete and repeated literature,unclear research results,and untrue and unreliable literature.Finally,87 eligible literatures were included.Results EZH2 can promote the growth,invasion,metastasis and metabolism of glioma cells.EZH2 can be used as the upstream gene of non-coding RNA such as miRNA and lncRNA to inhibit the expression of miRNA and lncRNA,thus promoting the occurrence and development of brain glioma.In addition,EZH2 can also be regulated by miRNA,lncRNA and circRNA,that is,EZH2 can also be used as a downstream target gene of non-coding RNA to regulate the malignant progression of glioma.Conclusion The complex regulatory network composed of EZH2 and non-coding RNA plays an important role in the occurrence and development of glioma,suggesting that EZH2 is a potential therapeutic target for glioma.Therefore,further study on the molecular mechanism and function of EZH2 in the occurrence and development of brain glioma will help to develop a new therapeutic scheme for brain glioma targeting EZH2,so as to improve the cure rate of brain glioma patients.
