细胞焦亡相关蛋白NLRP3、IL-1β、Caspase-1在溃疡性结肠炎黏膜中的表达及意义 被引量:3

摘要 目的 通过研究细胞焦亡相关蛋白NLRP3、IL-1β、Caspase-1在溃疡性结肠炎(ulcerative colitis, UC)患者结肠黏膜中的表达,比较其差异性,探讨其在UC发病机制中的作用及相关性。方法 取27例UC患者结肠黏膜标本,根据内镜及组织学进行分级,其中轻度13例,中重度14例。同时收集确诊为结直肠癌并行外科手术切除的结直肠断端正常的结肠黏膜标本25例。采用免疫组化SP法检测所有患者肠黏膜中NLRP3、IL-1β、Caspase-1的表达情况。结果 NLRP3、IL-1β、Caspase-1在UC患者结肠黏膜中的蛋白表达高于癌旁组织对照组,差异均有统计学意义(H=110.57,P<0.01)。IL-1β的表达与病理分级呈正相关(r=0.511,P<0.01)。NLRP3、Caspase-1蛋白表达与病理分级无相关性。结论 IL-1β与UC炎症的发生发展密切相关,NLRP3、Caspase-1参与炎症损伤,介导细胞焦亡,从而影响UC炎症的发生发展,提示细胞焦亡参与UC的发病过程。 Objective To study the expression of NLRP3, IL-1β and Caspase-1 in the colonic mucosa of patients with ulcerative colitis(UC), compare their differences, and to explore their role and correlation in the pathogenesis of UC.Methods Colonic mucosa specimens were collected from 27 UC patients and graded according to endoscopy and histology, including 13 mild cases and 14 moderate severe cases. Meanwhile, 25 colonic mucosal specimens were collected from normal controls who were diagnosed with colorectal cancer and underwent surgical resection. Immunohistochemical SP method was used to detect the expression of NLRP3, IL-1β and Caspase-1 in intestinal mucosa of all patients.Results The protein expressions of NLRP3, IL-1β and Caspase-1 in the colon mucosa of UC patients were significantly higher than those of the control(H=110.57, P<0.01). The expression of IL-1β was positively correlated with pathological grade(r=0.511, P<0.01). There was no correlation between NLRP3 and Caspase-1 protein expression and pathological grade.Conclusion IL-1β is closely related to the occurrence and development of UC inflammation. NLRP3 and Caspase-1 are involved in inflammatory injury and mediate pyroptosis, thus affecting the occurrence and development of UC inflammation, suggesting that pyroptosis is involved in the pathogenesis of UC.
