小白菊内酯、含笑内酯、(+)-Ainsliadimer A和大黄素对活化后肝星状细胞增殖能力的影响

摘要 目的探讨小白菊内酯、含笑内酯、(+)-Ainsliadimer A和大黄素对活化后肝星状细胞(hepatic stellate cells,HSC)增殖能力的影响。方法将大鼠HSC-T6细胞用含酒精的培养基诱导48 h使其活化;然后配置小白菊内酯、含笑内酯、(+)-Ainsliadimer A和大黄素不同浓度梯度含10%血清的DMEM培养基,分别培养活化后的HSC-T6,观察HSC-T6细胞的形态,以MTT法观察细胞增殖能力的变化。结果MTT结果显示,与对照组相比,小白菊内酯、含笑内酯、(+)-Ainsliadimer A和大黄素均可抑制活化后HSC的增殖(P<0.05),且抑制率呈浓度依赖关系。其中,(+)-Ainsliadimer A抑制细胞增殖的能力最强。结论小白菊内酯、含笑内酯、(+)-Ainsliadimer A和大黄素可能通过抑制活化的HSC增殖有效逆转酒精诱导的肝纤维化。 Objective To investigate the effects of parthenolide,micheliolide,(+)-Ainsliadimer A and emodin on the proliferation of activated hepatic stellate cells(HSC).Methods Rats HSC-T6 cells were induced with the alcohol-containing medium for 48 hours to activate them.Then,the activated HSC-T6 cells were cultured in DMEM medium containing 10%serum with different concentrations of parthenolide,micheliolide,(+)-Ainsliadimer A and emodin,respectively.The morphology of HSC-T6 cells was observed,and the proliferation of HSC-T6 cells was observed by MTT assay.Results The results of MTT showed that the proliferation of activated HSC was inhibited by parthenolide,micheliolide,(+)-Ainsliadimer A and emodin compared with the control group(P<0.05),and the inhibition rate was concentration-dependent.Among them,the ability of inhibiting the proliferation cell by using(+)-Ainsliadimer A was the best.Conclusion Parthenolide,micheliolide,(+)-Ainsliadimer A and emodin may effectively reverse alcohol-induced liver fibrosis by inhibiting the proliferation of activated HSC.
