湖北贝母内生真菌Curreya pityophila化学成分及其生物活性研究

摘要 采用硅胶柱色谱、Sephadex LH-20凝胶柱色谱、分析型和制备型HPLC的方法对湖北贝母内生真菌Curreya pityophila发酵液的乙酸乙酯层萃取物进行研究,共分离纯化得到9个化合物.通过波谱学数据分析并结合其物理性质,将其分别鉴定为:6-epi-5'-hydroxy-mycosporulone(1)、Massarigenin A(2)、Massarilactone-A acetonide(3)、trans-4-hydroxymellein(4)、2-Hydroxy-2,4-dimethyl-3(2H)-benzofuranone(5)、2,7-Dihydroxy-2,4-dimethyl-3(2H)-benzofuranone(6)、3-Carbomethoxy-4-methylphenol(7)、(+)-4,8-Dihydroxy-7-(2-hydroxy-ethyl)-6-methoxy-3,4-dihydro-2H-naphthalen-1-one(8)、Alternariol(9).以上化合物均为首次从Curreya pityophila中分离得到.采用MTT法对化合物1~9进行了人乳腺癌细胞(MCF-7)的细胞毒活性检测,结果显示这些化合物在40μM的浓度下均未表现出明显的抑制活性. By using gel column chromatography,sephadex LH-20,and preparative HPLC,nine compounds were isolated from the endophytic fungus Curreya pityophila of Fritillaria hupehensis.The structures of the compounds were determined to be 6-epi-5'-hydroxy-mycosporulone(1),Massarigenin A(2),Massarilactone A acetonide(3),trans-4-hydroxymellein(4),2-Hydroxy-2,4-dimethyl-3(2H)-benzofuranone(5),2,7-Dihydroxy-2,4-dimethyl-3(2H)-benzofuranone(6),3-Carbomethoxy-4-methylphenol(7),(+)-4,8-Dihydroxy-7-(2-hydroxy-ethyl)-6-methoxy-3,4-dihydro-2H-naphthalen-1-one(8),Alternariol(9).All compounds were isolated from the fermentation broth of Curreya pityophila for the first time.The cytotoxic activity of compounds 1-9 in human breast cancer cells(MCF-7)was tested by MTT assay,and the results showed that none of these compounds had significant inhibitory activity at a concentration of 40μM.
