基于新型冠状病毒基因型CT值变化规律指导疫情研判 被引量:1

摘要 目的 基于新型冠状病毒基因型CT值变化规律,建立指导疫情研判机制。方法 采用圣湘科技股份有限公司和中元汇吉生物技术股份有限公司生产的新型冠状病毒核酸检测试剂,分别对不同质量浓度梯度的4种疫苗、3种质控品进行双孔检测,对8个临床阳性样本进行单孔检测,记录O基因(ORF1ab)和N基因的CT值并进行相关分析。结果 非试剂阳性质控品、重组疫苗不能检测到新型冠状病毒基因型;灭活疫苗、第三方质控品O基因扩增效率高于N基因,即O基因CT值低于N基因,此情况刚好与临床阳性样本相反。结论 结合新型冠状病毒基因型CT值变化规律,有助于疫情研判,降低疫情流调过度反应。 Objective To establish a mechanism to guide the research and judgment of the epidemic situation based on the change rules of the computed tomography(CT) value of the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2) genotype. Methods SARS-CoV-2 nucleic acid detection reagents produced by Shengxiang Technology Co.,Ltd. and Zhongyuan Huiji Biotechnology Co.,Ltd. were used for double-hole detection on 4 vaccines and 3 quality control products with different concentration gradients,and single-hole detection on 8 clinical positive samples. CT values of ORF1ab(O gene) and N gene were recorded and analyzed. Results The SARS-CoV-2 genotype could not be detected in non-self-reagent positive quality control products and recombinant vaccines. The amplification efficiency of O gene was higher than that of N gene in inactivated vaccines,and third-party quality control products,that is,the CT value of O gene was lower than that of N gene,which was just the opposite of the results of clinical positive samples. Conclusion The change rules of CT value of SARS-CoV-2 genotype will help to study and judge the epidemic situation and reduce the overreaction to the epidemic.
