丁苯酞软胶囊对小鼠阿尔茨海默病学习记忆障碍的影响

Butylphthalide Soft Capsule Ameliorates Learning and Memory Impairment in Alzheimer′s Disease Mice Model by Inhibiting Neuronal Pyroptosis

目的研究丁苯酞软胶囊对阿尔茨海默病(alzheimer′s disease,AD)模型小鼠学习记忆障碍的影响。方法采用小鼠侧脑室内注射Aβ1-42制备模型模拟AD样症状,术后第2天开始灌胃给予丁苯酞,连续给药,第19天开始进行Y迷宫、Morris水迷宫行为学实验;TUNEL法细胞检测;试剂盒检测小鼠海马焦亡标志蛋白Caspase-1活性;Western Blot方法考察海马炎症小体NLRP3的表达。结果给予丁苯酞软胶囊治疗25 d可明显改善Aβ1-42所致小鼠AD样症状,减少神经元焦亡及炎症反应。结论丁苯酞可能会成为治疗AD的候选药物。

Objective To study the effects of dl-3-n butylphthalide(NBP)soft capsules on learning and memory impairment in Alzheimer′s disease(AD)model mice.Methods The mice were injected with Aβ1-42 into the lateral ventricle to induce AD-like symptoms.On the second day after operation,butylphthalide was given by gavage,and Y maze and Morris water maze were performed on the 19th day;cell detection by TUNEL method;the activity of the pyroptosis marker protein Caspase-1 in the hippocampus of mice was detected by kit;Western Blot was used to detect the expression of NLRP3 in the hippocampus.Results Butylphthalide soft capsule treatment for 25 days can significantly improve the AD-like symptoms induced by Aβ1-42,and reduce neuronal pyroptosis and inflammatory response in mice.Conclusion NBP may be a candidate for AD treatment.
