Faim2基因敲除增加小鼠心肌肥厚

摘要 目的建立Fas凋亡抑制分子2(Faim2)基因敲除小鼠,评价Faim2基因敲除对于小鼠主动脉弓缩窄(TAC)手术诱导的病理性心肌肥厚的影响。方法构建Faim2敲除的品系小鼠并繁殖,用PCR法和Western blot鉴定基因型;用主动脉弓缩窄手术建立小鼠心肌肥厚模型,并用苏木精-伊红以及天狼星红染色检测Faim2基因敲除对于小鼠心肌肥厚的影响。结果Faim2基因敲除小鼠的构建与繁殖均获得成功,Faim2基因敲除对于小鼠病理性心肌肥厚有促进的作用。结论Faim2基因敲除加重小鼠病理性心肌肥厚。 Objective To detect the effect of Faim2 gene knockout on cardiac hypertrophy induced by ransverse aortic constriction(TAC) in mice.Method Faim2 knockout mice were constructed,and the genotype was identified by PCR and Western blot.Cardiac hypertrophy model was established by TAC.The effects of Faim2 knockout on pathological cardiac hypertrophy were evaluated by hematoxylin-eosin and picrosirius red staining at 4 weeks after TAC surgery.Result Faim2 gene knockout mice were successfully generated.We found that Faim2 knockout could significantly exacerbate cardiac hypertrophy and fibrosis induced by TAC in mice.Conclusion Faim2 gene knockout aggravates cardiac hypertrophy in mice.
