摘要
目的探讨夜仁合有效部位改善果蝇睡眠作用的研究。方法以野生型Canton S品系黑腹果蝇为实验动物,分为空白组与给药组,分别给予浓度为0.25%、0.5%、1%、2%、4%的夜仁合水煎液和60%醇洗脱部位,以睡眠时间、睡眠—觉醒节律和自主活动次数为指标,用果蝇活动监测系统(DAMS)全天监测,研究夜仁合有效部位改善果蝇睡眠的作用。结果与空白组比较,夜仁合水煎液2%、4%剂量组可显著延长果蝇睡眠时间,减少自主活动次数(P<0.05);夜仁合60%醇洗脱部位1%、2%及4%剂量组可显著延长果蝇睡眠时间,减少自主活动次数(P<0.05或P<0.01)。结论夜仁合水煎液和60%醇洗脱部位均有镇静催眠作用,相比于水煎液,60%醇洗脱部位具有更显著改善果蝇睡眠的药理作用。
Objective To explore the effects of the effective parts of YERENHE on improving sleep of Drosophila.Methods Wild-type Canton S strain Drosophila were used as experimental animals and were divided into a blank group and an administration group.The YERENHE water decoction and 60%alcohol elution site with the concentration of 0.25%,0.5%,1%,2%,4%were treated in Drosophila,and the taking sleep time,sleep-wake rhythm and the number of autonomous activities were measured,the Drosophila Activity Monitoring System(DAMS)was used to monitor the whole day to study the effect of the effective parts of YERENHE on improving sleep of Drosophila.Results Compared with the blank group,the 2%and 4%dose groups of YERENHE water decoction group can significantly prolong the sleep time of Drosophila and reduce the number of autonomous activities(P<0.05).The 1%,2%and 4%dose groups of YERENHE 60%alcohol elution site can significantly prolong the sleep time of Drosophila and reduce the number of autonomous activities(P<0.05 or P<0.01).Conclusion YERENHE water decoction and 60%alcohol elution site had sedative and hypnotic effects,revealing a more obvious pharmacological effect in improving the sleep of Drosophila.
