子宫内膜癌组织中miR-195表达及其与预后的关系

Expression of mi R195-in endometrial carcinoma tissues and its relationship with prognosis

目的研究并分析子宫内膜癌(EC)组织中miR-195的表达水平及其与患者预后的关系。方法选取2014年2月至2015年5月于湖北医药学院附属东风医院就诊的126例EC患者,术后均取癌组织样本送病理检测。比较EC癌组织以及正常癌旁组织中miR-195的表达水平,分析miR-195与EC患者临床病理特征的关系,随访统计EC患者术后5年的生存情况,应用Logistic回归分析miR-195与EC患者预后生存的相关性。结果EC患者癌组织中miR-195相对表达量低于癌旁内膜组织(P<0.05);miR-195在不同肌层浸润深度、不同淋巴结转移、不同分化程度癌组织中的表达量差异均有统计学意义(P<0.05);随访至术后5年,126例EC患者中共有79例患者存活(62.70%)、47例患者死亡(37.30%),miR-195高表达患者5年生存率高于低表达患者(P<0.05);不同年龄、BMI、病理类型患者的5年生存率比较差异无统计学意义(P>0.05);肌层浸润深度≥1/2、发生淋巴结转移、低分化、miR-195低表达患者的5年生存率低于肌层浸润深度<1/2、未发生淋巴结转移、中高分化和miR-195高表达患者(P<0.05);Logistic回归分析结果显示,肌层浸润深度[OR=3.428(95%CI:1.023~11.488)]和淋巴结转移[OR=2.214(95%CI:1.039~4.719)]均为影响患者预后生存的独立危险因素,高分化[OR=0.601(95%CI:0.374~0.966)]和miR-195高表达[OR=0.713(95%CI:0.535~0.951)]均为影响患者预后生存的保护因素(P<0.05)。结论EC患者癌组织中miR-195较癌旁内膜组织呈现低表达,其作为患者预后生存的保护因素,与患者临床病理特征表现以及预后生存结局密切相关。

Objective To study and analyze the expression level of mi R-195 in endometrial carcinoma(EC)tissues and its relationship with prognosis of patients.Methods A retrospective analysis was performed on the 126 patients with EC treated in Dongfeng Hospital Affiliated to Hubei University of Medicine between February 2014 and May 2015.The samples of carcinoma tissues were tested after surgery.The expression level of mi R-195 in EC tissues and normal para-carcinoma tissues was compared.The relationship between mi R-195 and clinicopathological characteristics in EC patients was analyzed.The survival status of EC patients at 5 years after surgery was statistically analyzed by follow-up.The correlation between mi R-195 and prognosis of EC patients was analyzed by Logistic regression analysis.Results The relative expression level of mi R-195 in EC tissues was lower than that in para-carcinoma tissues(P<0.05).There were significant differences in expression level of mi R-195 among patients with different muscular infiltration depth,lymph node metastasis and differentiation degree(P<0.05).All patients were followed up for 5 years after surgery.In the 126 EC patients,there were 79 survival cases(62.70%)and 47 death cases(37.30%).The 5-year survival rate in high-expression mi R-195 patients was higher than that in low-expression patients(P<0.05).The difference in 5-year survival rate among patients with different age,BMI and pathological types was not statistically significant(P>0.05).The 5-year survival rate in patients with muscular infiltration depth≥1/2,lymph node metastasis,low differentiation and low expression of mi R-195 was lower than that in patients with muscular infiltration depth<1/2,no lymph node metastasis,moderate to high differentiation and high expression of mi R-195(P<0.05).The results of Logistic regression analysis showed that muscular infiltration depth[OR=3.428(95%CI:1.023-11.488)]and lymph node metastasis[OR=2.214(95%CI:1.039-4.719)]were independent risk factors of prognosis,while high differentiation[OR=0.601(95%CI:0.374-0.966)]and high expression of mi R-195[OR=0.713(95%CI:0.535-0.951)]were protective factors(P<0.05).Conclusions The expression level of mi R-195 in EC tissues is lower than that in para-carcinoma tissues.As a protective factor of prognosis,mi R-195 is closely related to clinicopathological characteristics and prognosis of patients.
