狂犬病病毒GX074株P蛋白48~78位区域氨基酸联合M蛋白突变株的构建及特性研究

摘要 为探究广西狂犬病病毒街毒GX074株P蛋白联合M蛋白对生物学特性的影响,实验以构建的弱毒疫苗株rRC-HL感染性cDNA克隆质粒pRC-HL为基础,将GX074株P蛋白的48~78位区域氨基酸及M蛋白联合替换到pRC-HL相同位置,进行突变病毒的拯救,经间接免疫荧光实验及序列测定获得突变体rRC-HL(GX074P 48~78 M)。结果表明,突变体病毒rRC-HL(GX074P 48~78 M)基因组稳定,拯救后的荧光灶小于亲本弱毒rRC-HL株,大于亲本强毒GX074株及CVS-11株;在细胞中的复制能力表现出与rRC-HL株相似的特点,多步生长曲线显示突变体病毒滴度略低于rRC-HL,而稍高于GX074株及CVS-11株。说明突变体毒株rRC-HL(GX074P 48~78 M)相较于亲本弱毒rRC-HL株复制能力有所降低,比亲本强毒GX074株复制能力略高。
