芪天扶正胶囊的HPLC指纹图谱研究

Study on the HPLC fingerprint of Qitianfuzheng Capsules

目的 建立芪天扶正胶囊的高效液相色谱(high performance liquid chromatography, HPLC)指纹图谱,为其质量控制提供依据。方法 用Shimadzu C18柱(250 mm×4.6 mm, 5μm)色谱柱,以乙腈-1 mL·L^(-1)甲酸水溶液为流动相梯度洗脱,流速为0.8 mL·min^(-1),柱温为25℃,检测波长为258 nm。结果 10批芪天扶正胶囊的HPLC指纹图谱有35个共有指纹峰,相似度均大于0.9,通过与混合对照品比对,指认出19种化学成分:腺苷、没食子酸、5-羟甲基糠醛、红景天苷、酪醇、莫诺苷、马钱苷、当药苷、毛蕊异黄酮苷、芦丁、木犀草苷、特女贞苷、芹菜苷、橙皮苷、橄榄苦苷、芒柄花苷、女贞苷G13、毛蕊异黄酮、木犀草素。结论 所建立的HPLC指纹图谱能够表达芪天扶正胶囊中多组分的整体特征,可用于其质量控制。

Objective To establish high performance liquid chromatography(HPLC) fingerprint of Qitianfuzheng Capsules, and provide basis for quality control.Methods The chromatographic separation was performed on a Shimadzu C18column(250 mm×4.6 mm, 5 μm) with acetonitrile-1 mL·L^(-1)formic acid as mobile phase at a flow rate of 0.8 mL·min^(-1). The column temperature was 25 ℃ and the detection wavelength was 258 nm.Results 10 batches of Qitianfuzheng Capsules had 35 common peaks in the HPLC chromatogram, and the similarity was greater than 0.9. By comparing with the mixed reference substances, 19 chemical components were identified, which were adenosine, gallic acid, 5-hydroxymethylfurfural, salidroside, tyrosol, morroniside, loganin, swertimarin, calycosin-7-glucoside, rutinum, luteoloside, specnuezhenide, apiin, hesperidin, oleuropein, ononin, nuezhenoside G13, calycosin and luteolin.Conclusion The established HPLC fingerprint can express the overall characteristics of multiple components in Qitianfuzheng Capsules, which can be used for the quality control.
