基于网络药理学探讨飞扬肠胃炎胶囊治疗胃溃疡的作用机制

摘要 目的基于网络药理学探讨飞扬肠胃炎胶囊治疗胃溃疡的作用机制。方法采用文献法检索收集飞扬肠胃炎胶囊的化合物成分,并通过SwissADME对成分进行筛选。使用SwissTargetPrediction数据库对化合物成分进行靶点预测。借助GeneCards和OMIM数据库获取胃溃疡相关靶点;采用String数据库构建蛋白质-蛋白质相互作用(PPI)网络图;通过DAVID数据库进行基因本体(GO)富集分析和京都基因和基因组百科全书(KEGG)通路分析,并采用Cytoscape3.8.0软件构建“飞扬肠胃炎胶囊-成分-靶点-胃溃疡”网络图。结果通过GeneCards、OMIM数据库检索到4958个胃溃疡相关靶点基因,取交集得到飞扬肠胃炎胶囊-胃溃疡交互靶点205个。共筛选出12个主要化合物成分,301个胃溃疡疾病靶点。网络分析结果表明,飞扬肠胃炎胶囊治疗胃溃疡可能与蛋白质磷酸化、信号转导、对药物的反应、凋亡过程的负调控、蛋白质自磷酸化等生物学过程及PI3K-Akt、cAMP、AMPK、VEGF等信号通路有关。结论飞扬肠胃胶囊治疗胃溃疡通过多个信号通路协同发挥作用,明确相关作用机制可为研究飞扬肠胃炎胶囊治疗胃溃疡提供可靠的理论指导。 Objective To explore the action mechanism of Feiyang Changweiyan capsules in treating gastric ulcer based on network pharmacology.Methods The components of Feiyang Changweiyan capsules were retrieved and collected by literature method,and the components were screened by SwissADME.The SwissTargetPrediction database was used to predict the target of active components.The related targets of gastric ulcer were obtained by GeneCards and OMIM database;the String database was used to construct protein-protein interaction(PPI)network diagram;the Gene Ontology(GO)enrichment analysis and the Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were performed by DAVID database,and Cytoscape3.8.0 software was used to construct the network diagram of"Feiyang Changweiyan capsules-component-target-gastric ulcer".Results Through Gene Cards and OMIM database,4958 gastric ulcer related target genes were retrieved,and 205 Feiyang Changweiyan capsules-gastric ulcer interactive targets were obtained by intersection.Twelve main components and 301 gastric ulcer disease targets were screened.The results of network analysis showed that Feiyang Changweiyan capsules in the treatment of gastric ulcer may be related to protein phosphorylation,signal transduction,response to drug,negative regulation of apoptosis process,protein autophosphorylation and other biological processes and PI3K-Akt,cAMP,AMPK,VEGF and other signaling pathways.Conclusion Feiyang Changweiyan capsules plays a synergistic role in the treatment of gastric ulcer through multiple signaling pathways,and clarifying the relevant mechanism of action can provide reliable theoretical guidance for the study of Feiyang Changweiyan capsules in the treatment of gastric ulcer.
