Pan-cancer analysis of SOX4 gene expression signature

目的通过生物信息学方法探讨并验证SOX4基因在泛癌中的表达特征。方法通过癌症基因组图谱(TCGA)和基因表达综合数据库(GEO)对SOX4基因进行泛癌分析,从基因表达、生存分析、基因突变、免疫浸润及富集分析等多个方面挖掘SOX4在不同肿瘤致病中的作用。利用定量逆转录聚合酶链反应(RT-qPCR)和蛋白质印迹法(Western blotting)验证SOX4在3种常见肿瘤细胞系中的表达情况。结果SOX4在大多数肿瘤中高表达,且与部分肿瘤患者的预后存在明显相关性。在口腔鳞状细胞癌细胞、乳腺癌细胞和人非小细胞肺癌细胞中,SOX4 mRNA和蛋白的水平明显升高。在膀胱尿路上皮癌患者中,SOX4的突变发生率最高。在部分肿瘤中,SOX4的表达与免疫细胞浸润密切相关。SOX4可能通过“病毒致癌”和“肿瘤MicroRNA”通路参与肿瘤的发生。结论SOX4在多种肿瘤的致病中起重要作用,可能是治疗肿瘤的有效靶点。

Objective To explore and verify the expression characteristics of the SOX4 gene in pan-cancer through bioinformatics methods.Methods The TCGA and GEO databases were used to conduct the pan-cancer analysis of the SOX4 gene.The role of the SOX4 gene in the pathogenesis of different tumors was explored from the aspects of gene expression,survival analysis,gene variation,immune infiltration,and enrichment analysis.The expression of the SOX4 gene in three common tumor cell lines was verified by RT-qPCR and Western blotting.Results SOX4 gene was highly expressed in most tumors,and it had a clear correlation with the prognosis of some tumor patients.The levels of SOX4 mRNA and protein were significantly increased in SCC-4,MCF-7 and A549 cell lines.SOX4 gene had the highest mutation rate in bladder urothelial carcinoma patients.In some tumors,the expression of the SOX4 gene was closely related to immune cell infiltration.SOX4 might be involved in tumorigenesis through“viral carcinogenesis”and“tumor microRNA”pathways.Conclusion SOX4 gene plays an important role in the pathogenesis of various tumors and may be a target for effective tumor therapy.
