大叶茜草素诱导黑色素瘤B16F10凋亡 被引量:1

The Effect of Alizarin Macrophylla on Apoptosis of Melanoma B16F10 Cells

摘要 目的研究大叶茜草素对黑色素瘤B16F10细胞的作用机制。方法使用MTT法(其中大叶茜草素浓度为0、40、80、160和320μmol/L)、平板克隆(其中大叶茜草素浓度为0、10、20和40μmol/L)来检测B16F10的增殖能力,用划痕实验(其中大叶茜草素的浓度为0、75、150和300μmol/L)来检测B16F10的迁移能力,使用凋亡与坏死检测试剂盒(其中大叶茜草素的浓度为0、75、150和300μmol/L)来检测大叶茜草素是否诱导B16F10凋亡,使用蛋白免疫印迹法(其中大叶茜草素的浓度为0、75、150和300μmol/L)来检测B16F10细胞中MTA1基因蛋白水平的表达。结果大叶茜草素可剂量依赖性抑制B16F10细胞的增殖和迁移,诱导B16F10凋亡;经大叶茜草素处理后B16F10细胞中MTA1具有下调趋势,且呈剂量依赖性。结论大叶茜草素可以通过下调MTA1来抑制B16F10细胞的增殖、迁移并诱导凋亡。 Objective To investigate the effect of alizarin macrophylla on melanoma B16F10 cells.Methods MTT assay and plate cloning formation assay were used to detect the viability of alizarin macrophylla at different concentrations on B16F10 cells,scratch test was used to detect the migration,immunofluorescence staining was used to detect the apoptosis,and western blot was used to determine protein expression.Results Alizarin macrophylla inhibited the proliferation,cell clone formation and migration of B16F10 cells in a dose-dependent manner,induced the apoptosis of B16F10 cells,and down-regulated the expression of MTA1 in B16F10 cells.Conclusion Alizarin macrophylla can inhibit the proliferation,migration and induce apoptosis of B16F10 cells by down-regulating MTA1 expression.
