摘要 已有研究表明TLP(Tubby-like protein)蛋白在植物应对非生物胁迫方面具有重要作用。本研究利用生物信息学方法在白菜中鉴定得到14个TLP家族基因,并对其理化性质、基因结构特征、蛋白保守性、系统进化关系、组织表达模式及对盐胁迫的响应情况进行了初步分析。结果表明,该家族基因分布在白菜10条染色体中的7条。蛋白分子量介于38 735.69~52 747.34 D,等电点介于9.16~9.88。基因结构分析表明,有1个TLP基因含有9个CDS,绝大多数基因CDS为4~5个。亚细胞定位预测结果表明,该基因家族成员主要定位在细胞核、线粒体和细胞质基质。启动子顺式作用元件分析表明,该基因家族成员含有包括光响应元件、脱落酸应答元件、厌氧反应应答元件在内的大量逆境胁迫应答元件。组织特异性表达分析发现大多数BrTLPs在花中表达量较高,在茎、叶片和果荚中的表达量与根相似;绝大多数白菜TLP基因对盐胁迫都有不同程度的响应,其中BrTLP14对盐胁迫反应最强烈,暗示该基因可能在白菜响应盐胁迫过程中发挥重要作用。 Studies have shown that TLP(Tubby-like protein) family plays critical roles in plant response to abiotic stresses. In this study, 14 members of the TLP family were identified from Brassica rapa by bioinformatics methods, and their physicochemical properties, gene structure, protein conservation, phylogenetic relationship, tissue expression patterns, and response to salt stress were analyzed. The results showed that the gene family was located on 7 out of 10 chromosomes of Brassica rapa. The molecular weight of the proteins ranged from 38 735.69 to 52 747.34 D, and the isoelectric point ranged from 9.16 to 9.88. Gene structure analysis showed that one gene had 9 CDS, and most genes had 4 ~ 5 CDS. The prediction of subcellular localization showed that the gene family members were mainly located in nucleus, mitochondria and cytoplasmic matrix. The analysis of promoter cis-acting elements showed that the gene family members contained a large number of stress response elements including light response elements, abscisic acid response elements and anaerobic response elements. Tissue-specific expression analysis showed that most BrTLPs were highly expressed in Brassica rapa flowers, and the expression level in stems, leaves and pods were similar to that in roots. Most of the BrTLP genes in Brassica rapa had different responses to salt stress, and BrTLP14 showed the strongest response to salt stress, suggesting that BrTLP14 might play an important role during response to salt tolerance in Brassica rapa.
