羊传染性脓疱灭活疫苗的制备及临床免疫效果观察 被引量:2

摘要 为制备羊传染性脓疱灭活疫苗并评价其免疫效果,试验中制备羔羊睾丸原代细胞后增殖羊传染性脓疱病毒并运用切向流超滤膜对病毒液进行10倍浓缩,使用β-丙内酯灭活的病毒液按1:1混合206佐剂制备羊传染性脓疱灭活疫苗,在常发羊传染性脓疱的羊场进行免疫试验并观察免疫效果。羔羊睾丸细胞在接种病毒后有明显细胞病变,经毒力检测病毒原液滴度为10^(6.3)TCID50/0.1 mL,10倍浓缩液滴度为10^(7.5)TCID_(50)/0.1 m L,用制备的羊传染性脓疱灭活疫苗对孕羊及羔羊采用肌内注射的方式进行免疫,经观察灭活疫苗对羔羊有明显保护力。切向流超滤膜浓缩效果良好,对保留病毒粒子,提高病毒浓度效果显著;羊传染性脓疱灭活疫苗对常发病羊场羔羊保护效果显著。 In order to prepare the inactivated goat infectious pustular vaccine and evaluate its immune effect,the primary cells of the testis of lambs were prepared to proliferate goat infectious pustular virus followed by the virus solution was concentrated 10 times with tangential flow ultrafiltration membrane.The inactivated goat infectious pustular vaccine was prepared with β-propiolactone inactivated virus with 206 adjuvant at 1:1.The immune test was carried out in the goat farm where goat infectious pustules often occurred,and the immune effect was observed.The testicular cells of lambs had obvious cytopathic changes after inoculation of the virus.The titer of the origin virus solution was 10^(6.3)TCID_(50)/0.1mL,and the titer of 10 times concentrated solution was 10^(7.5)TCID_(50)/0.1mL.Pregnant goat and lambs were immunized by intramuscular injection with the prepared inactivated goat infectious pustule vaccine.It was observed that the inactivated vaccine had obvious protective effect on lambs.Tangential flow ultrafiltration membrane had a good concentration effect,and has a significant effect on retaining virus particles and increasing virus concentration.Inactivated goat infectious pustule vaccine had a significant protective effect on lambs in the common infected goat farms.
