凝血因子Ⅷ和凝血因子Ⅸ活性检测影响因素的分析

摘要 目的 验证离心力对凝血因子FⅧ(FⅧ)活性和凝血因子FⅨ(FⅨ)活性检测的影响。方法 用全自动血凝分析仪测定室温700 g、1500 g、396 g和4℃1000 g、1500 g离心15 min以及4℃2700 g离心10 min,不同离心力条件下即刻和冷冻后48 h FⅧ和FⅨ的活性并进行数据统计分析。结果 6种不同离心方法得到的血浆,不管是立即检测还是冻存后检测,FⅧ和FⅨ的活性水平在不同离心力条件下没有显著性差异(P>0.05),同一离心力条件下冷冻后FⅧ活性水平跟即刻检测结果有统计学差异(P<0.05),同一离心条件下FⅨ活性在即刻和冷冻后检测结果无统计学差异(P>0.05)。结论 从离心机类型、离心力转速到离心时间和离心温度多方面分析,在一定离心力范围内不同离心力对FⅧ活性和FⅨ活性检测的影响不大,冻融会影响凝血因子Ⅷ活性水平检测,凝血因子Ⅸ活性检测不受冻融影响。 Objective To verify the effect of centrifugal force on coagulation factor Ⅷ activity and coagulation factor Ⅸ activity detection.Methods The activity of coagulation factor Ⅷ and coagulation factorⅨ was measured immediately and 48 hours after freezing under different centrifugal force conditions(Room temperature 700 g,1500 g,396 g and 4 ℃ 1000 g,1500 g for 15 minutes and 4 ℃ 2700 g for 10 minutes) by automatic coagulation analyzer and the data was statistically analyzed.Results There was no significant difference in the activity levels of coagulation factor Ⅷ and coagulation factor Ⅸ in plasma obtained by different centrifugation methods under different centrifugal force conditions,either immediately or after freezing(P> 0. 05). The coagulation factor Ⅷ activity level after freezing under the same centrifugal force was statistically different from the immediate detection results(P<0.05),there was no significant difference between the activity of coagulation factor Ⅸ immediately and after freezing under the same centrifugal condition(P>0.05).Conclusion From the analysis of centrifuge type,centrifugal speed,centrifugal time and centrifugal temperature,within a certain range of centrifugal force,different centrifugal forces have little influence on the activity test of coagulation factor Ⅷ and coagulation factor Ⅸ. Freeze-thaw can affect the activity level test of coagulation factor Ⅷ,and the activity test of coagulation factor Ⅸ can not be affected by freeze-thaw.
