重度子痫前期孕妇胎盘中miR-296-3p和MMP9的表达及临床意义 被引量:1

摘要 目的探讨miR-296-3p和基质金属蛋白酶9(MMP9)在重度子痫前期(sPE)孕妇胎盘组织中的表达及临床意义。方法选取2021年1月至2022年1月在亳州市人民医院住院并剖宫产分娩的sPE孕妇52例为sPE组,另选取正常妊娠剖宫产分娩的孕妇52例作为对照组。应用逆转录聚合酶链反应(RT-qPCR)检测两组孕妇胎盘组织中miR-296-3p和MMP9 mRNA的表达情况,蛋白质免疫印迹(Western-blot)检测MMP9蛋白的表达水平。采用Pearson法分析miR-296-3p与MMP9的相关性,以及两者与sPE孕妇的临床特征和妊娠结局的关系,双荧光素酶报告系统检测miR-296-3p和MMP9的靶向关系。结果sPE组miR-296-3p相对表达量高于对照组[(0.72±0.10)比(0.35±0.07)],sPE组MMP9 mRNA和蛋白表达均低于对照组[(0.21±0.06)比(0.57±0.10),(0.16±0.06)比(0.65±0.08)],差异有统计学意义(t=22.336、22.360、-35.602,P均<0.001);miR-296-3p和MMP9 mRNA的表达呈负相关(P<0.05);sPE胎盘组织中miR-296-3p表达与收缩压和舒张压呈正相关(P均<0.05),与新生儿出生体质量和分娩孕周呈负相关(P均<0.05),sPE胎盘组织中MMP9 mRNA的表达与收缩压和舒张压呈负相关(P均<0.05),与新生儿出生体质量和分娩孕周呈正相关(P均<0.05)。荧光素酶报告基因实验证实MMP9是miR-296-3p靶基因。结论miR-296-3p高表达、MMP9低表达与sPE的严重程度和预后相关,推测miR-296-3p可能通过负调控MMP9的表达参与sPE的发生发展。
