摘要
目的:明确朝鲜苍术对脾虚大鼠的药理学指标影响,以北苍术作为阳性药,并就实验结果对比阳性药组对朝鲜苍术的健脾作用进行评价,为朝鲜苍术更合理的使用提供实验依据。方法:将40只雄性SD大鼠随机分为空白组、模型组、阳性药组、朝鲜苍术组四组,每组10只。用空白组外的3组采用饥饱失常、疲劳过度加苦寒泻下的复合因素法造模21天建立脾虚模型。造模第22-28天,阳性药组与朝鲜苍术组大鼠分别用等剂量的北苍术与朝鲜苍术给药干预。称量各组大鼠的体质量、胸腺质量和脾脏质量,计算胸腺指数和脾脏指数,酶联免疫吸附法(ELISA)测定各组大鼠血清中淀粉酶(AMS)、胃泌素(GAS)、胃动素(MTL)、胆囊收缩素(CCK)、饥饿素(Ghrelin)、生长抑素(SS)、免疫球蛋白G(IgG)、干细胞因子(SCF)、白细胞介素4(IL-4)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)、γ干扰素(IFN-γ)、血管活性肠肽(VIP)、表皮生长因子(EGF)及三叶因子1(TFF1)的水平变化情况。结果:与空白组相比较,模型组大鼠的体质量、胸腺指数、脾脏指数显著下降(P<0.05),血清中AMS、IFN-γ、IgG、IL-4和TFF1水平显著下降(P<0.05),CCK、GAS、Ghrelin、MTL、SS、VIP、SCF、EGF、IL-6和TNF-α水平显著上升(P<0.05)。与模型组相比较,阳性药组大鼠血清中SS和IL-4水平显著回调(P<0.05),IL-6水平未呈现差异性;朝鲜苍术组大鼠血清中,除SS水平呈现回调趋势但未呈现显著性,IL-4和IL-6水平未表现出差异性外,其余各组指标皆呈现回调趋势,其趋势均呈现显著性(P<0.05)。与阳性药组相比较,朝鲜苍术组大鼠血清中MTL、EGF、IFN-γ、TNF-α水平显著升高(P<0.05),CCK、AMS、Ghrelin、IgG水平显著降低(P<0.05)。结论:阳性药与朝鲜苍术对脾虚都具有良好的调节作用,在大鼠脾虚引发的体质量变化以及胸腺、脾脏和代偿机制的改善方面,朝鲜苍术表现更优;在对脾虚大鼠的免疫调节、炎症反应调节以及溃疡的修复方面,阳性药表现更优。
Objective:To clarify the influence of the Atractylodes coreana indicators on spleen deficiency rats, take Atractybodes chinensis as a positive medicine, and evaluate the spleen effect of the Atractylodes coreana compared to the experimental results, and provide more reasonable use of Atractylodes coreana to provide more reasonable use Experimental basis.Methods:40 male SD rats were randomly divided into four groups of blank groups, model groups, positive pharmaceutical groups, and Atractylodes coreana group, each group includes 10 rats.The three groups other than the blank group were used for 21 days to establish the spleen deficiency model by using the compound factor method of starvation and satiety disorder, excessive fatigue and bitter cold diarrhea.On the 22nd-28th day of modeling, the rats in the positive drug group and the Atractylodes coreana group were intervened with the same doses of Atractybodes chinensis and Atractylodes coreana respectively.The body weight, thymus weight and spleen weight of the rats in each group were weighed, and the thymus index and spleen index were calculated.The levels of Amylase(AMS),Gastrin(GAS),Motilin(MTL),Cholecystokinin(CCK),Ghrelin(Ghrelin),Somatostatin(SS),Immunoglobulin G(IgG),Stem Cell Factors(SCF),Interleukin 4(IL-4),Interleukin 6(IL-6),Tumor Necrosis Factor Alpha(TNF-α),Interferon Gamma(IFN-γ),Vasoactive Intestinal Peptide(VIP),epidermal growth factor(EGF) and trefoil factor 1(TFF1) in serum of rats in each group were determined by enzyme-linked immunosorbent assay(ELISA).Results:Compared with the blank group, the body weight, thymus index, and spleen index of the model group were significantly decreased(P<0.05),the levels of AMS,IFN-γ,IgG,IL-4 and TFF1 in serum decreased significantly(P<0.05),and the levels of CCK,GAS,Ghrelin, MTL,SS,VIP,SCF,EGF,IL-6 and TNF-α increased significantly.Compared with the model group, the serum levels of SS and IL-4 in the positive drug group were significantly reversed(P<0.05),but the IL-6 level did not show any difference;In the serum of the rats in the Atractylodes coreana Rhizoma group, except for SS,which showed a downward trend but no significance, and no difference in the levels of IL-4 and IL-6,the other indexes showed a downward trend, and the trends were all significant(P<0.05).Compared with the positive drug group, the serum levels of MTL,EGF,IFN-γ and TNF-α in the Atractylodes coreana group were significantly increased(P<0.05),and the levels of CCK,AMS,Ghrelin and IgG were significantly decreased(P<0.05).Conclusion:Both positive medicine and Atractylodes coreana have a good regulating effect on spleen deficiency, and Atractylodes coreana performs better in the improvement of body weight, thymus, spleen and compensatory mechanism caused by spleen deficiency in rats;in terms of immune regulation, inflammatory response regulation and ulcer repair in rats with spleen deficiency, the positive drugs perform better.
