摘要
目的:基于网络药理学探讨半枝莲-龙葵药对治疗结直肠癌(CRC)的潜在作用机制。方法:利用TCMSP筛选出半枝莲-龙葵有效成分并预测靶点;运用GeneCards检索出CRC相关靶点;通过STRING建立药物与疾病交集靶点的PPI网络模型,运用Metascape对靶点进行GO和KEGG富集分析。结果:得到药物有效成分33种、靶点230个,CRC靶点1 341个,半枝莲-龙葵治疗CRC的主要成分为槲皮素、木犀草素、汉黄芩素、β-谷甾醇、黄芩素,作用的核心靶点为PTGS2、AKT1、HSP90AB1、PTGS1、BCL2,主要参与的生物学通路为PI3K-Akt信号通路、NF-κB信号通路、HIF-1信号通路、FoxO信号通路等。结论:半枝莲-龙葵的成分槲皮素、木犀草素、汉黄芩素可能作用于PTGS2、AKT1、HSP90AB1、PTGS1、BCL2等靶点,通过PI3K-Akt信号通路、NF-κB信号通路、HIF-1信号通路等发挥治疗CRC的作用。
Objective:To explore the potential mechanism of Scutellaria barbata and Solanum nigrum in the treatment of colorectal cancer based on network pharmacology.Methods:TCMSP was used to screen the effective components of Scutellaria barbata and Solanum nigrum and predict the target;The CRC related targets were retrieved by GeneCards;The PPI network model of drug and disease intersection target was established by STRING,and the target was enriched and analyzed by go and KEGG by Metascape.Results:33 kinds of effective components, 230 targets and 1 341 CRC targets were obtained.The main components of Scutellaria barbata longkui in the treatment of CRC were quercetin, luteolin, wogonin β-sitosterol and baicalein, PTGS2,AKT1,HSP90AB1,PTGS1 and BCL2 are core targets.The main biological pathways involved PI3K Akt signaling pathway and NF-κB signal pathway, HIF-1 signal pathway, FoxO signal pathway, etc.Conclusion:Quercetin, luteolin and Scutellaria baicalensis may act on targets such as PTGS2,AKT1,HSP90AB1,PTGS1 and BCL2 through PI3K-Akt signaling pathway and NF-κB signaling pathway and HIF-1 signaling pathway play a role in the treatment of CRC.
