高通量测序分析新疆不同产区葡萄酒对大鼠肠道菌群多样性的影响

Effect of wine from different regions in Xinjiang on intestinal flora diversity in rats based on high-throughput sequencing technology

摘要 将雄性SD大鼠随机分为5组(A1~A5),分别灌胃新疆焉耆盆地、伊犁河谷、天山北麓、吐哈盆地葡萄酒、蒸馏水8周后,收集粪便,采用高通量测序技术分析大鼠肠道菌群变化,探讨不同产区葡萄酒对大鼠肠道菌群的影响。结果表明,各组大鼠肠道菌群具有一定的差异,主要共有优势菌门为厚壁菌门(Firmicutes)和拟杆菌门(Bacteroidetes),优势菌属为乳杆菌属(Lactobacillus),A1~A4组中主要优势菌门和菌属的相对丰度之和均比A5组显著升高(P<0.05),其中A2组升高幅度最大,其有害菌门变形菌门(Proteobacteria)、脱硫菌门(Desulfobacterota)相对丰度显著降低(P<0.05);有益菌属罗姆布茨菌属(Romboutsia)、艾克蔓菌属(Akkermansia)及乳杆菌属(Lactobacillus)相对丰度显著增加(P<0.05)。结合多级物种差异判别分析,A2组肠乳杆菌(Lactobacillus intestinalis)的富集最具差异性(P<0.05)。说明适量摄入伊犁河谷干红葡萄酒一定程度上能改善肠道菌群。 Male SD rats were randomly divided into five groups(A1-A5).After 8 weeks of gavage with wine of Yanqi basin,Yili River Valley,northern foothill of Tianshan mountain and Tuha basin in Xinjiang and distilled water,the feces were collected,the changes of intestinal flora in rats were analyzed by high-throughput sequencing technology,and the effects of wine from different producing areas on intestinal flora of rats were discussed.The results showed that the intestinal flora of rats in each group was different.The main common dominant bacteria phylum were Firmicutes and Bacteroidetes,the dominant bacteria genus was Lactobacillus,and the sum of relative abundance of dominant bacteria phylum and genus in group A1-A4 was significantly higher than that of group A5(P<0.05),and the increase of group A2 was the largest,and the relative abundance of harmful bacteria phylum Proteobacteria and Desulfobacterota was significantly decreased(P<0.05).The relative abundance of beneficial bacteria genera Romboutsia,Akkermansia and Lactobacillus increased significantly(P<0.05).Combining with difference discriminant analysis of multilevel species,the enrichment of L.intestinalis in group A2 was the most different(P<0.05).It showed that moderate intake of dry red wine from Yili River Valley could improve intestinal flora to some extent.
