榆荚仁黄酮对慢性睡眠剥夺小鼠抗氧化能力的影响 被引量:3

摘要 本试验旨在探究榆荚仁黄酮对睡眠剥夺小鼠的抗氧化作用。将42只4周龄无特定病原体(SPF)级雄性ICR小鼠随机分为7组,每组6只。使用改良多水平台睡眠剥夺法构建慢性睡眠剥夺模型,造模成功后灌胃28 d。低、中、高浓度榆荚仁黄酮组每天分别灌胃30、60、120 mg/kg的榆荚仁黄酮,空白对照组(不再进行睡眠剥夺)灌胃等量的生理盐水,阳性对照组灌胃等量的褪黑素,模型对照组灌胃等量的生理盐水,羧甲基纤维素钠(CMC-Na)对照组灌胃等量的0.5%CMC-Na溶液。结果表明:1)低、中、高浓度榆荚仁黄酮组大脑和肝脏中血红素氧合酶-1(HO-1)、还原型辅酶Ⅰ(Ⅱ)醌氧化还原酶1(NQO1)活性均显著高于模型对照组和CMC-Na对照组(P<0.05),低、中浓度榆荚仁黄酮组肠道中HO-1、NQO1活性均显著高于模型对照组和CMC-Na对照组(P<0.05)。模型对照组和CMC-Na对照组血清8-羟基脱氧鸟苷(8-OH-dG)含量显著高于其他各组(P<0.05)。2)阳性对照组及低、中、高浓度榆荚仁黄酮组大脑、肝脏中核因子E2相关因子2(Nrf2)、NQO1和HO-1 mRNA相对表达量均显著高于空白对照组、模型对照组和CMC-Na对照组(P<0.05),阳性对照组及中、高浓度榆荚仁黄酮组肠道Nrf2、NQO1和HO-1 mRNA相对表达量均显著高于模型对照组和CMC-Na对照组(P<0.05)。综上所述,榆荚仁黄酮可提高慢性睡眠剥夺小鼠大脑、肝脏、肠道中HO-1、NQO1活性和Nrf2、HO-1、NQO1 mRNA相对表达量,改善慢性睡眠剥夺小鼠氧化应激损伤。 This experiment was conducted to investigate the effects of elm pod kernel flavone on antioxidant capacity of chronic sleep deprived mice.Forty-two 4-week-old specific pathogen free(SPF)grade male ICR mice were randomly divided into 7 groups with 6 mice per group.A chronic sleep deprivation model was constructed using the modified multiple platforms method,and gavaged for 4 weeks after successful modeling.Mice in low,medium and high concentration flavone groups were gavaged with 30,60 and 120 mg/kg elm pod kernel flavone per day,respectively,mice in the blank control group(no sleep deprivation)were gavaged with the same amount of distilled water,mice in the positive control group were gavaged with the same amount of melatonin,mice in the model control group were gavaged with the same amount of distilled water,and mice in the sodium carboxymethyl cellulose(CMC-Na)control group were gavaged with the same amount of 0.5%CMC-Na solution.The results showed as follows:1)the activities of heme oxygenase-1(HO-1)and NAD(P)H:quinine oxidoreductase 1(NQO1)in brain and liver of low,medium and high concentration flavone groups were significantly higher than those of model control group and CMC-Na control group(P<0.05),and the activities of HO-1 and NQO1 in intestine of low and medium concentration flavone groups were significantly higher than those of model control group and CMC-Na control group(P<0.05).The serum 8-hydroxy-deoxyguanosine content of model control group and CMC-Na control group was significantly higher than that of other groups(P<0.05).2)The mRNA relative expression levels of nuclear factor erythroid-2 related factor 2(Nrf2),NQO1 and HO-1 in brain and liver of positive control group and low,medium and high concentration flavone groups were significantly higher than those of blank control group,model control group and CMC-Na control group(P<0.05),and the mRNA relative expression levels of Nrf2,NQO1 and HO-1 in intestine of positive control group and medium,high concentration flavone groups were significantly higher than those of model control group and CMC-Na control group(P<0.05).In conclusion,the elm pod kernel flavone can improve the activities of HO-1 and NQO1 and mRNA relative expression levels of Nrf2,HO-1 and NQO1 in brain,liver and intestine of chronic sleep deprived mice,and ameliorate oxidative stress injury of chronic sleep deprived mice.
