摘要
目的通过生物信息学的方法对DPP4基因与蛋白进行系统的分析,为DPP4在病毒侵入宿主细胞和连花清瘟胶囊等药物防治等方面的研究打下生物信息学基础。方法首先分析DPP4蛋白的亲疏水性、信号肽结构、跨膜结构等基本性质,再通过SOPMA、SWISS-MODEL、STRING、MEGA-X等软件分析蛋白质的结构、相互作用关系、进化关系等高级属性进行分析,并通过其性质和属性分析DPP4在病毒侵入和药物治疗原理方面的作用。结果DPP4是一个亲水性非经典分泌蛋白,具有一个跨膜结构,并在小肠中表达较为丰富,在细胞中主要分布在内质网。DPP4的翻译后修饰位点较多,三级结构呈较为特殊的哑铃形,蛋白主要涉及糖代谢调控、炎性反应以及病毒侵入宿主细胞等生物过程。人类DPP4蛋白与啮齿类动物DPP4蛋白的亲缘性较差。结论连花清瘟胶囊对DPP4具有明确的靶向作用,并且DPP4蛋白与病毒侵入人体密切相关,因此其对病毒性肺炎的防治效果是可靠且明确的。
Objective The systematic analysis of DPP4 gene and protein through bioinformatics methods laid a bioinformatics foundation for research on viruses invading host cells and Lianhua Qingwen Capsules and other drugs for epidemic prevention and control.Methods First,analyzed the basic properties of DPP4 protein,such as hydrophilicity and hydrophobicity,signal peptide structure,and transmembrane structure,and then analyzed the structure,interaction relationship,evolution relationship,and other advanced properties of the protein through software such as SOPMA,SWISSMODEL,STRING,MEGA-X,etc.Finally,analyzed the role of DPP4 in virus invasion and drug treatment principles through these properties and attributes.Results DPP4 was a hydrophilic non-classical secreted protein with a transmembrane structure and was abundantly expressed in the small intestine.It was mainly distributed in the endoplasmic reticulum in cells.DPP4 had many post-translational modification sites,and the tertiary structure was a relatively special dumbbell shape.The DPP4 protein was mainly involved in biological processes such as the regulation of sugar metabolism,inflammatory response,and virus invasion of host cells.Human DPP4 protein had poor affinity with rodent DPP4 protein.Conclusion The DPP4 protein was closely related to the invasion of the virusinto the human body.Since Lianhua Qingwen Capsule had a clear targeting effect on DPP4,its prevention and treatment effect on viral pneumoniais reliable and clear.
