摘要
以聚己内酯-聚乙二醇-聚己内酯为原料,通过静电喷雾的方法,制备了具有缓释作用的载万古霉素微球.探究了浓度、电压、温度、加药量4个参数对微球性质的影响,并进行了微球的模拟体外释放实验,确定最佳参数为纺丝液质量百分比为15%、电压14 kV、温度35℃,加药量1 g万古霉素、4 g聚乙二醇、6 g乙醇,载药量高达(11.03±0.52)%,包封量高达(88.12±1.26)%,缓释效果可达一月以上.
Using polycaprolactone-polyethylene glycol-polycaprolactone as raw materials,the microspheres containing vancomycin with sustained release effect were prepared by electrostatic spraying.In this experiment,the influence of the four parameters of concentration,voltage,temperature,and dosage on the properties of microspheres was explored,and a simulated invitro release experiment of microspheres was carried out.The optimal parameters were determined as the concentration of 15wt%,voltage of 14 kV,and temperature.At 35℃,the dosage is 1 g Van+4 g PEG+6 g EtOH,the drug loading is as high as 11.03±0.52%,the encapsulation is as high as 88.12±1.26%,and the sustained-release effect can reach more than one month.
