摘要
目的 探究高危型人乳头瘤病毒(HPV)感染宫颈病变患者外周血环鸟嘌呤核苷酸腺嘌呤核苷酸合成酶/干扰素激活蛋白/TANK结合激酶1(cGAS/STING/TBK1)信号通路基因相对表达量。方法 选取2015年4月-2021年4月中部战区总医院妇产科收治的高危型HPV感染宫颈病变患者89例为研究组,同期无HPV感染的宫颈病变患者89例为对照组,采用第2代杂交捕获实验检测高危型HPV DNA载量,荧光定量聚合酶链反应(PCR)检测外周血cGAS、STING和TBK1 mRNA表达水平。结果 研究组外周血单个核细胞中cGAS、STING和TBK1 mRNA水平高于对照组(P<0.05);不同高危型HPV DNA载量患者外周血cGAS/STING/TBK1信号通路基因表达的比较差异有统计学意义(P<0.05),其中高载量组cGAS、STING和TBK1 mRNA水平最高,低载量组最低(P<0.05);不同高危型HPV感染类型患者外周血cGAS/STING/TBK1信号通路基因表达的比较差异无统计学意义。结论 高危型HPV感染对宫颈病变患者外周血cGAS/STING/TBK1信号通路基因表达影响较大,高危型HPV DNA载量与其表达水平密切相关。
OBJECTIVE To explore the relative expression levels of cyclic guanine nucleotide adenine nucleotide synthase/stimulator of interferon genes/TANK binds kinase 1(cGAS/STING/TBK1) signaling pathways genes in peripheral blood of patients with high-risk human papillomavirus(HPV) infection and cervical lesions. METHODS A total of 89 patients with high-risk HPV infection and cervical lesions who were treated in General Hospital of the Central Theater Command from Apr 2015 to Apr 2021 were assigned as the study group, meanwhile, 89 cervical lesion patients who did not have HPV infection were chosen as the control group. The high-risk HPV-DNA load was detected by the second-generation hybrid capture test, the expression levels of peripheral blood cGAS, STING and TBK1 mRNA were detected by means of fluorescent quantitative polymerase chain reaction(PCR). RESULTS The expression levels of cGAS, STING and TBK1 mRNA in mononuclear cells of peripheral blood were higher in the study group than in the control group(P<0.05). There were significant differences in the expression levels of peripheral blood cGAS/STING/TBK1 signaling pathways genes among the patients with different loads of high-risk HPV-DNA(P<0.05), the expression levels of cGAS, STING and TBK1 mRNA were the highest in the high-load group, the lowest in the low-load group(P<0.05). There were no significant differences in the expression levels of peripheral blood cGAS/STING/TBK1 signaling pathways genes among the patients with different types of high-risk HPV infection. CONCLUSION The high-risk HPV infection has great effect on the expressions of cGAS/STING/TBK1 signaling pathways genes in peripheral blood of the patients with cervical lesions, and the high-risk HPV-DNA load is closely associated with the expression levels of cGAS/STING/TBK1 signaling pathways genes.
