美洲大蠊提取物CⅡ-3调控p53蛋白抑制K562细胞增殖

Inhibition of K562 Cell Proliferation by p53 Protein Regulated by Periplaneta americana Extract CⅡ-3

目的:探讨美洲大蠊提取物CⅡ-3对人白血病细胞K562增殖的影响。方法:以不同浓度的美洲大蠊提取物CⅡ-3作用于K562细胞,CCK-8法检测其对细胞增殖的影响,集落形成实验检测其对细胞增殖能力的影响,流式细胞术检测K562细胞的细胞周期,蛋白质印迹法检测细胞中磷酸化p53蛋白的表达。结果:美洲大蠊提取物CⅡ-3可抑制K562细胞增殖,其最佳作用浓度为80μg/mL,作用时间72 h;CⅡ-3能降低K562细胞集落形成率;使阻滞在G0/G1期的K562细胞比例增高,S期的细胞比例减少;促进磷酸化p53蛋白表达上调。结论:CⅡ-3通过调控p53蛋白抑制K562细胞增殖。

Objective:To investigate the effect of Periplaneta americana extract CⅡ-3 on the proliferation of human leukemia cell K562.Methods:K562 cells were treated with different concentrations of Periplaneta americana extract CⅡ-3,and the effects of CⅡ-3 on the proliferation of K562 cells were detected by CCK-8 assay.Colony formation assay was used to detect its effect on proliferation of K562 cells.The cell cycle of K562 cells was detected by flow cytometry.The expression of phosphorylated p53 protein was detected by Western blot.Results:Periplaneta americana extract CⅡ-3 could inhibit the proliferation of K562 cells,and the optimal concentration and duration of action were 80μg/mL and 72 h respectively.Periplaneta americana extract CⅡ-3 reduced the colony formation rate of K562 cells.The proportion of K562 cells blocked in G0/G1 phase increased,while that in S phase decreased.The expression of phosphorylated p53 protein was up-regulated.Conclusion:Periplaneta americana extract CⅡ-3inhibits the proliferation of K562 cells by regulating p53 protein.
