摘要
双硫仑作为FDA批准的戒酒药在临床上被广泛使用。双硫仑主要通过抑制NLRP3炎症小体活化、Gasdermin D蛋白膜孔形成、中性粒细胞胞外捕网的形成及释放、抑制核转录因子κB激活等机制,抑制炎症反应并改善炎性相关疾病。此外,双硫仑与其他药物联合使用可提升其抗炎作用。现对双硫仑抗炎作用的分子机制及其在炎性疾病中的作用进行综述,以期为双硫仑作为抗炎药物的应用提供参考。
Disulfiram approved by FDA has a long history of being used for alcohol abuse.This agent inhibits inflammatory response and improves inflammation-related diseases mainly by inhibiting the activation of NLRP3 inflammasome, the formation of Gasdermin D membrane pores, the formation and release of Netrophic extracelluar traps, and activation of NF-κB.In addition, disulfiram combined with other drugs can enhance its anti-inflammatory effect.This paper reviews briefly the major findings concerning the molecular mechanism of disulfiram in connection with inflammatory diseases, hoping to provide the basis for the application and research of disulfiram as an anti-inflammatory drug.
