麒麟丸对卵巢颗粒细胞损伤的修复作用研究

摘要 【目的】在细胞水平探讨麒麟丸对卵巢储备功能下降的治疗作用及机制。【方法】培养人卵巢颗粒细胞KGN,采用雷公藤多苷诱导建立KGN细胞损伤模型,给予麒麟丸水提物干预,采用四甲基偶氮唑盐(MTT)法观察细胞增殖情况,Western Blot法检测细胞抗苗勒激素(AMH)、Smad2、Smad3蛋白表达水平,酶联免疫吸附分析(ELISA)检测细胞上清雌二醇(E2)水平。【结果】2.5~10 mg·L^(-1)麒麟丸均能促进受损KGN细胞的增殖活性(P<0.05),上调AMH、Smad3蛋白表达,提高E2分泌水平(P<0.01);2.5 mg·L^(-1)麒麟丸可促进Smad2蛋白表达水平(P<0.05)。【结论】麒麟丸对卵巢储备功能下降治疗作用可能是通过Smads信号通路保护卵巢颗粒细胞实现的。 Objective To investigate the therapeutic effect and mechanism of Qilin Pills on reduced ovarian reserve function at the cellular level.Methods KGN cells,a human ovarian granulosa cell line,were cultured and induced by tripterygium glycosides to establish the KGN cell injury model.Water extracts of Qilin Pills were given for intervention.The cell proliferation was observed by MTT method,and the protein expression levels of antimüllerian hormone(AMH),Smad2 and Smad3 in cells were detected by Western Blot.The level of estradiol(E2)in cell supernatant was detected by enzyme-linked immunosorbent assay(ELISA).Results All 2.5~10 mg·L^(-1)Qilin Pills promoted the proliferative activity of damaged KGN cells(P<0.05),up-regulated AMH and Smad3protein expression and increased E2secretion level(P<0.01);2.5 mg·L^(-1)Qilin Pills promoted Smad2 protein expression level(P<0.05).Conclusion The therapeutic effect of Qilin Pills on reduced ovarian reserve function may be achieved through the protection of ovarian granulosa cells by the Smads signaling pathway.
